作者范现成(2019)在《毒害艾美耳球虫感染雏鸡小肠lncRNAs、miRNAs和CircRNAs表达谱分析》一文中研究指出:毒害艾美耳球虫(Eimeria necatrix)是鸡球虫病中主要致病虫种之一,常寄生于鸡小肠和盲肠的上皮细胞内,导致受损肠道严重出血,病鸡常表现腹泻、血痢、生长缓慢等临床症状,有较高发病率和死亡率,对鸡的养殖造成严重威胁。目前,该病的防治主要依赖药物和疫苗,药物虽然具有较好的驱虫效果,但易产生药物残留和耐药性;疫苗在鸡球虫的预防控制中可有效避免化学药物带来的许多副作用,但其存在高成本、易散毒、免疫保护力不够等缺点。近些年来的研究表明,非编码RNA(non-coding RNA,ncRNA)可在宿主与寄生虫的相互作用中发挥重要的调节作用,特别是长链非编码RNA(long non-coding RNA,lncRNA)、环状RNA(Circular RNA,CircRNA)和微小RNA(microRNA,miRNA)参与了许多疾病的发生和发展过程,并已作为多种疾病防治的潜在靶标和诊断、预后的生物标志。基于此,本研究以E.necatrix感染雏鸡为模型,利用高通量测序技术系统研究与E.necatrix感染相关的宿主小肠的全转录组表达谱。1.获得了E.necatrix感染雏鸡小肠的mRNA和lncRNA表达谱:将8000个E.necatrix卵囊经口感染10 d的雏鸡,在感染后108 h取雏鸡小肠中段进行RNA-seq分析,共获得1543个显著差异表达的mRNA(707个上调mRNA和836个下调mRNA)和95个显著差异表达的lncRNA(49个上调lncRNA和46个下调lncRNA)。共表达网络分析显示,这些lncRNA和mRNA之间存在37134条共表达关系,其中38个lncRNA顺式调节73个mRNA,而87个lncRNA反式调节1453个mRNA。GO功能和KEGG通路富集分析发现,差异表达的lncRNA的靶基因显著富集于E.necatrix感染相关的AMPK、PPAR和JAK-STAT等免疫通路。2.获得了E.necatrix感染雏鸡小肠的miRNA表达谱:在雏鸡小肠中段组织中鉴定出1266个miRNA,包括663个已知的miRNA和603个新的miRNA。进一步研究发现,E.necatrix感染引起了雏鸡小肠35个miRNA显著差异表达(16个上调和19个下调);功能分析发现其中的21个miRNA的4568个靶mRNA显著富集于2047个GO条目(转录、转录后调控和粘附分子连接等)和54个KEGG通路(钙信号通路、MAPK信号通路和胰岛素信号通路等)。3.获得了E.necatrix感染雏鸡小肠的CircRNA表达谱:在雏鸡小肠中段组织中共鉴定出4153个CircRNA序列,E.necatrix感染引起了雏鸡小肠13个CircRNA显著差异表达(9个上调CircRNA和4个下调CircRNA)。功能分析发现,这些CircRNAs来源基因主要参与正调控NF-κB信号通路、磷脂酰肌醇信号通路等。CircRNA-miRNAmRNA互做网络分析发现,本研究中差异表达的CircRNAs可能通过吸附2个差异表达的miRNA,间接调控62个差异表达的靶mRNA,而这些靶mRNAs主要富集在MAPK、JAK-STAT等通路。综上所述,本研究运用RNA-seq技术对E.necatrix感染雏鸡的小肠中段组织进行测序分析发现,E.necatrix感染改变了小肠中的mRNAs、lncRNAs、miRNAs和CircRNAs表达谱,功能分析发现这些RNA分子可能参与了E.necatrix与雏鸡小肠的互作过程。这些研究结果为深入揭示E.necatrix与雏鸡的互作机制提供了基础数据。
