作者刘石刚(2019)在《基于胺基荧光纳米探针的分子识别新策略及其生化分析应用研究》一文中研究指出:发展性能优异的分析方法在生物医药分析、食品质量和安全控制、环境监测和法医鉴定等领域非常重要。荧光分析具有灵敏度高、操作简便和成本经济等优点,在许多领域有着广泛的应用。荧光探针是荧光分析的重要工具,近年来,各类新型荧光探针的制备及其在生化分析中的应用受到了研究者的广泛关注和探索。纳米技术的发展为各个学科的发展带来了新的视角和进步,同样的,荧光纳米探针的开发和应用为荧光分析技术提供了新的机遇,例如,荧光纳米探针在荧光成像、生化传感、医学诊断和治疗等方面得到了越来越多的研究和应用,促进了各学科的快速融合以及新理论和新技术的出现和发展。胺为含氮有机物,可以看作是氨的烃基衍生物。胺分子中氮原子上具有未共用的电子对,所以胺类物质一般给电子能力强,也易与水形成氢键,水溶性好。很多胺类化合物是维持生命活动的重要物质,因此基于胺制备的许多材料具有良好的生物相容性。以胺化合物为前驱体制备发光纳米材料时,氮原子作为掺杂元素或者给电子能力强的氨基作为荧光助色单元可以提高材料的光学性能;同时,制备的纳米材料表面会存在氨基,这有利于其作为探针时对目标分析物的特异性,也可以增强材料的水溶性和便于材料表面修饰和功能化,这些都能提高荧光探针的性能和分析表现。本论文主要以胺类化合物(聚乙烯亚胺、三聚氰胺和乙二胺)为主要前体物质,利用不同的材料制备方法(如交联反应、自组装、热裂解、液相剥离和水热法等),制备了几种发光纳米材料,包括荧光聚合物纳米颗粒、石墨相氮化碳纳米片和荧光碳点,通过电镜、光谱和表面分析等表征技术,研究了它们的化学和光学性质;进一步的,以这些发光纳米材料作为荧光探针,结合分子光谱分析技术,构建离子和分子的识别新方法,探究分析方法的性能和机理,实现了对几种痕量目标物的特异性和高效检测。本论文的主要研究内容总结如下:1.基于聚乙烯亚胺-甲醛荧光纳米颗粒的铜离子探针及IMPLICATION逻辑门的构建基于聚乙烯亚胺(PEI)交联甲醛制备了一种发射蓝色荧光的水溶性聚合物纳米颗粒(PEI-F NPs),并以PEI-F NPs作为荧光探针实现对铜离子的灵敏性检测。相较于共轭聚合物纳米颗粒和其它的一些荧光纳米材料,PEI-F NPs的制备相对简便且环境友好。PEI-F NPs具有本质的荧光发射,并且对铜离子表现出灵敏性地响应。在pH 4.0的HAc-NaAc缓冲溶液中,PEI-F NPs的荧光能够被铜离子灵敏且特异性地猝灭,其它离子几乎没有干扰。基于此现象,构建了铜离子荧光传感器,该传感器性能良好,检出限达到62 nM。进一步探讨了荧光识别机理:铜离子是通过和PEI-F NPs表面的功能基团络合实现从PEI-F NPs到铜离子的电子转移,进而导致PEI-F NPs的荧光猝灭。另外,发现半胱氨酸能够恢复PEI-F NPs-Cu2+体系的荧光,且荧光的“开-关-开”具有可逆性,基于这种现象,本章还成功构建了一个IMPLICATION逻辑门。2.聚乙烯亚胺-葡萄糖荧光纳米颗粒的制备、光学性质及其在苦味酸检测中的应用利用PEI和葡萄糖通过席夫碱反应和自组装制备了一种具有强荧光发射的水溶性非共轭聚合物纳米颗粒(PEI-G NPs)。这种发光材料的制备方法简便且条件温和。运用电镜和分子光谱等表征手段,探究了PEI-G NPs的形成机理、基本性质(例如微观形貌、浓度依赖和溶剂依赖的荧光)和荧光起源。同时发现PEI-G NPs的荧光能够被硝基化合物苦味酸(PA)灵敏地猝灭,而其他硝基爆炸物和结构相似的化合物干扰较小。因此,使用PEI-G NPs作为荧光探针,构建了水溶液中PA快速、灵敏和选择性检测的新方法。该方法性能优异,操作简便,经济环保,对PA的检测有较宽的线性范围(0.05-70μM),检出限低至26 nM。进一步的,本章还探究了PA引发PEI-G NPs荧光猝灭的可能机制,归因于有效的能量转移、静态猝灭及内滤效应的综合作用。运用该方法于实际样品中的PA检测,所得结果相对标准偏差(RSD)较低,加标回收率满意。3.基于胺基荧光聚合物纳米颗粒的比色和荧光双信号响应的pH探针及多逻辑门运算本章利用PEI和水杨醛通过亚胺交联和自组装制备了一种非传统共轭聚合物荧光纳米颗粒(PEI-S NPs)。该荧光纳米颗粒的制备方法环境友好且操作简便。利用透射电镜、核磁共振、FT-IR光谱、热重分析等技术对PEI-S NPs进行了表征。研究了PEI-S NPs的光学性质,发现PEI-S NPs的荧光和紫外-可见吸收都表现出对pH快速、灵敏和可逆地响应。pH响应的机理被详细探究:双光学信号pH响应归结于PEI-S NPs表面的对质子具有响应的两种官能团(酚羟基和氨基);pH依赖的荧光归因于电子转移,由氨基的质子化程度控制;而pH依赖的吸收则由酚羟基的解离控制。比色法和荧光法均获得宽pH响应线性范围,因此,PEI-S NPs可作为纳米探针用于水性介质中的pH传感。本章中还基于PEI-S NPs的pH依赖的双光学信号变化构建了多个逻辑门,包括双输出的INHIBIT逻辑门。4.基于g-C3N4/CoOOH纳米体系构建比率荧光传感平台用于碱性磷酸酶活性分析以三聚氰胺为前驱体通过高温热解法和液相剥离法合成了具有蓝色荧光的石墨相氮化碳(g-C3N4)纳米片,并以g-C3N4纳米片复合羟基氧化钴(CoOOH)纳米片,基于目标物启动的竞争性氧化还原反应实现了碱性磷酸酶(ALP)活性的比率荧光分析。分析原理如下:在g-C3N4/CoOOH纳米体系溶液中,邻苯二胺(OPD)被氧化,所得氧化产物(OxOPD)发出橙色荧光;同时,g-C3N4的蓝色荧光被OxOPD猝灭。由于抗坏血酸(AA)和CoOOH之间特异性的氧化还原反应,AA可导致CoOOH纳米片的降解。因此,在AA存在下,CoOOH纳米片优先被AA还原破坏,而OPD难以被氧化成OxOPD,在这种情况下,g-C3N4发出强蓝色荧光而来自OxOPD的橙色荧光很弱。AA诱导的双信号变化使得比率测定ALP成为可能,因为ALP可以催化L-抗坏血酸-2-磷酸酯(AAP)水解产生AA。因此,基于g-C3N4/CoOOH纳米体系和OPD,以AAP为底物可以实现ALP活性的比率荧光分析,检出限达0.92 U L-1。在此比率分析系统中,g-C3N4纳米片和OxOPD作为信号读出单元,CoOOH纳米片作为传感识别单元。此方法应用于人血清样品中ALP活性的分析获得了较好的效果。5.基于荧光联用散射构建比率分析新策略:以CDs/CoOOH纳米体系检测抗坏血酸为例本章以PEI和柠檬酸为前体物质通过水热法合成了荧光性能优异的碳点(CDs)。目前的比率分析虽然能获得两个或多个信号,但往往是同一种光或电信号。纳米材料具有尺寸和形貌依赖的物理化学性质,比如电导、紫外可见吸收和荧光等,毫无疑问,合理的纳米体系可以实现荧光和光散射的同时响应。在此原理基础上,本章提出了一种新的比率分析策略:利用两种不同的光学信号——荧光和散射设计比率传感器。基于荧光、光散射和衍射的原理,提出了两种信号收集策略用于在相同激发光下同时获得荧光和散射信号,并实现比率测定。一种是收集普通(下转换)荧光和二级散射(SOS)信号,另一种是记录二级衍射光激发的荧光(SODL-荧光)和一级散射(FOS)或倍频散射(FDS)信号。作为概念验证实验,使用制备的荧光碳点和羟基氧化钴纳米片(CDs/CoOOH)体系对抗坏血酸(AA)进行了两种模式的比率测定。除了构建比率光学传感器之外,荧光联用散射还可以作为一种新技术用于监测聚集诱导的荧光猝灭或增强。本章提出的新型比率分析模式为纳米材料的应用和生化分析提供了新思路。6.基于刻蚀银纳米颗粒实现荧光-二级散射比率分析双氧水、葡萄糖和氧化酶活性以乙二胺和柠檬酸为前体物质水热法合成了一种荧光碳点(CDs),并通过同时利用银纳米颗粒(AgNPs)的荧光猝灭能力和高光散射能力构建了过氧化氢(H2O2)及其相关分析物的比率分析新方法。以CDs/AgNPs纳米复合体系作为传感系统,通过同时收集荧光和二级散射(SOS)信号来实现H2O2的比率检测。在CDs/AgNPs系统中,一方面,由于共振能量转移作用,CDs的荧光会被AgNPs猝灭,而H2O2的引入,AgNPs会被刻蚀,使CDs的荧光恢复;另一方面,随着AgNPs的分解,纳米体系的散射会显著降低。也就是说,刻蚀AgNPs会增强荧光并降低散射,即CDs/AgNPs系统对H2O2具有双信号响应。通过同时收集荧光和SOS信号,实现了灵敏地比率测定H2O2,检出限为0.86μM。CDs/AgNPs系统对H2O2的比率测定提供了涉及H2O2的通用分析平台:检测反应后会产生H2O2的分析物(例如,葡萄糖、胆碱和尿酸等)。作为模型试验,应用分析体系对葡萄糖和葡萄糖氧化酶(GOx)活性进行了比率测定。
