作者唐小飞(2019)在《西藏热泉嗜热真菌分离鉴定、产酶及生物活性初步研究》一文中研究指出:木聚糖酶是糖苷水解酶家族中的一个重要门类,与纤维素酶一起构成了近25%的酶类市场。该酶主要应用于纸浆造纸、食品和饲料等行业的漂白。在工业生产中,首先要考虑的是木聚糖酶的耐热性问题。嗜热真菌是一类能够在高温环境下生存的特殊真菌类群,近年来,被发现能够产生木聚糖酶等多种热稳定酶和活性代谢产物而成为研究的热点。本研究以西藏热泉为研究对象,分离嗜热真菌资源。研究结果如下:1.利用4种培养基,采用稀释涂布平板法分离西藏热泉真菌,并对其进行种属鉴定。从西藏热泉中共分离得到41株真菌,结合形态学特征和ITS序列分析对所分离的菌株进行初步鉴定,将其鉴定为四种嗜热真菌(Malbranchea cinnamomea,Thermomyces lauginosus,Thermomyces dupontii,Melanocarpus albomyces)和一种耐热真菌(Aspergillus fumigatus)。其中Melanocarpus albomyces尚未报道。2.通过高通量测序技术探究西藏热泉真核生物多样性,结果表明上述样品中的真核微生物主要归为6个门,19个纲,52个目,86个科,110个属。其中真菌界的优势属为曲霉属(Aspergillus)、枝孢属(Cladosporium)、Microbotryozyma、Ophiocordyceps、青霉属(Penicillium)、Rhizophydium、木霉属(Trichoderma)、Pyrenochaetopsis.说明西藏热泉真核微生物具有丰富的多样性。3.利用5种产酶筛选培养基通过透明圈的有无、大小,对5种真菌菌株的功能酶活性进行初步筛选。结果表明3种真菌产生淀粉酶,2种产生纤维素酶,5种产生木聚糖酶,1种产生蛋白酶,没有菌株产生糖苷酶。同时,对菌株THN8 Melanocarpus albomyces进行产木聚糖酶复筛,其酶活较高达到245.9U/mL。4.对从西藏热泉筛选的嗜热真菌Melanocarpus albomyces进行产木聚糖酶条件研究,确定产木聚糖酶的最佳培养基配方为玉米芯粉20 g/L,胰蛋白胨15 g/L,MgSO4 0.3 g/L,KH2PO4 0.35 g/L,最佳发酵条件为45℃,发酵时间为5 d,初始pH为5.5,接种量为8%(v/v)。优化后的木聚糖酶活比优化前提高了2.4倍。5.采用滤纸片法对分离得到的嗜热真菌发酵产物进行了抑菌试验及对球囊线蚓的抑制作用,其中Aspergillus fumigatus的发酵产物对金黄色葡萄球菌抑制作用效果最好,抑菌圈直径达到22 mm,其次为Malbranchea cinnamomea、Thermomyces dupontii抑菌圈直径达到20 mm。Thermomyces lauginosus的发酵产物对枯草芽孢杆菌的抑菌圈直径达到12 mm。Aspergillus fumigatus、Malbranchea cinnamomea、Thermomyces dupontii的发酵液对球囊线蚓有较强的杀伤作用。
