作者刘杰(2019)在《鹅细小病毒与鸭圆环病毒共感染增强致病性研究》一文中研究指出:鹅细小病毒(GPV)属细小病毒科(parvoviridae)、细小病毒属(parvovirus),主要引起小鹅瘟,近年来,其在鸭群中可引起鸭大舌短喙综合征。鸭圆环病毒(DuCV)感染可导致宿主免疫抑制,通常与其他病原体共同感染使疾病进展更加复杂。GPV与DuCV共感染在田间广泛存在,但目前对GPV与DuCV共感染对宿主的影响情况不明。为探究GPV和DuCV是否协同增强了致病性,本研究通过临床病例分离鉴定了GPV与DuCV;通过建立GPV与DuCV共感染的动物模型,以病毒学、病理学、分子生物学等方法进行了GPV与DuCV协同复制与协同致病研究。病毒分离鉴定结果显示,分离得到的GPV与DuCV均为当前田间流行株,并制备了GPV多克隆抗体和DuCV多克隆抗体用于实验检测。将GPV与DuCV腹腔接种于2日龄樱桃谷肉鸭,在感染后第10天、第20天和第30天进行剖杀。GPV与DuCV共感染鸭表现精神沉郁,生长发育迟缓,羽毛凌乱,体重增长显著缓慢。血涂片观察可见共感染组鸭血液中异嗜性粒细胞和淋巴细胞明显增多。对血清中各种酶活力指标进行检测,结果发现,感染前期共感染组谷丙转氨酶(ALT)水平显著高于单感染组和正常组,反映共感染GPV和DuCV对肝脏的损伤加重。共感染组的大体病变明显比单感染组严重,表现为肝脏出血坏死,脾脏萎缩苍白,胸腺、法氏囊严重萎缩,骨髓苍白。组织病理学变化表现为:免疫系统中,脾脏红髓和白髓边界消失,淋巴细胞稀疏,出血,异嗜性粒细胞浸润;胸腺淋巴细胞稀疏,胸腺小叶间大量异嗜性粒细胞浸润,胸腺小体崩解;法氏囊髓质淋巴细胞稀疏;骨髓造血细胞大量流失。消化系统中,十二指肠绒毛上皮坏死脱落;肝脏严重空泡变性并有炎性细胞浸润。泌尿系统中,肾脏出血,肾小管上皮细胞空泡变性、坏死脱落。通过qPCR对各个时间点不同组织内病毒载量进行检测,我们发现,感染初期共感染组的病毒载量显著低于单感染组,但随着病程的发展,单感染组的病毒载量下降,GPV和DuCV共感染动物体内两种病毒的载量明显升高,均显著高于单感染组。通过IHC检测GPV和DuCV在体内的分布,在肝脏、脾脏、肾脏、十二指肠、法氏囊内检测到阳性细胞。随着日龄的增加,共感染组阳性细胞数量及信号强度明显高于单感染组,但两种病毒分布定位基本一致。感染初期,两种病毒均定位在肝细胞核,接毒后第30天,肝细胞的胞浆和胞核均能检测到阳性信号。肾脏的髓袢处的肾小管可检测到阳性细胞。感染前期,十二指肠中的阳性细胞主要在肠隐窝处,后期存在于十二指肠肠绒毛的杯状细胞中。法氏囊中DuCV阳性细胞主要分布在髓质,GPV阳性细胞在皮质和髓质均可检测到。综上所述,GPV与DuCV可协同促进各自的复制、协同增强致病性。本研究从致病性和病毒复制两方面揭示了共感染的GPV与DuCV具有协同致病作用。本研究不仅对后续研究GPV与DuCV共感染机制明确了方向,更将为免疫抑制病或其它禽病共感染研究提供了科学依据。
Abstract
