作者周惠强(2019)在《大刺鳅(Mastacembelus armatus)繁殖生物学研究》一文中研究指出:大刺鳅(Mastacembelus armatus)属于合鳃目(Symbranchiformes)刺鳅科(Mastacembelidae)刺鳅属(Mastacembelus),大刺鳅肉质鲜美,营养丰富,价格昂贵,有一定的市场价值。但是近年来环境恶化加上渔民大肆捕捞,其野生资源逐渐减少。大刺鳅人工繁殖技术还不太成熟,对于大刺鳅的资源保护就变得更加重要,不仅要保护好大刺鳅的栖息环境,还要提升大刺鳅人工繁殖和增殖技术。本研究以大刺鳅为研究对象,对大刺鳅雌雄个体形态差异、繁殖生物学、血清性类固醇激素水平变化进行研究,同时克隆与性别相关的Foxl2基因,探究其在繁殖周期中的表达变化。本研究的主要结果如下:1.运用差异显著性分析、主成分分析、R-聚类分析和判别分析测量分析了248尾珠江流域野生大刺鳅3个计量性状和25个标准化形态性状。差异显著性分析结果表明,在方差均呈齐性(P>0.05)的性状中,下颚长/头长和L1-3/体长这两个标准化性状达到显著水平,12个性状达到极显著水平。在主成分分析中,提取7个主成分,累积贡献率达到77.040%,结合主成分分析和R-聚类分析的结果,表明大刺鳅个体的差异主要集中在头部、鱼体形态和尾部3个方面。通过逐步判别分析法从248尾大刺鳅的25个标准化形态性状中筛选出16个性状,建立性别判别方程,对雌、雄群体的综合判别准确率为69.2%。雌性比雄性头部和躯干部较长、眼间距较宽以及体高较高;而雄性比雌性上下颚较长、尾部较长和体长较长。上述研究结果可为大刺鳅性别鉴定提供科学方法和理论依据。2.本研究采用形态测量、组织学和酶联免疫测定等方法,对247尾大刺鳅的性腺发育规律和血清性类固醇激素的变化情况进行研究。在繁殖季节,大刺鳅雌鱼体色较淡呈浅黄色,腹部膨大,摸起来比较柔软,体形比较短粗;雄鱼体色较深呈暗黑色,腹部没有明显膨大,体形比较修长。大刺鳅的繁殖期主要集中在3-7月份,属于多次产卵,分批非同步类型,大刺鳅的精巢属于叶状型。大刺鳅的绝对繁殖力为455-7064粒,相对繁殖力为9.10-61.81粒/g。Pearson相关性检验表明,性腺成熟系数与肝脏指数相关性不显著(P>0.05),但和肥满度呈显著正相关关系(P<0.05),肥满度和肝脏指数呈显著负相关关系(P<0.05)。血清性类固醇激素水平的变化与性腺成熟系数的变化基本相似,在性腺成熟的繁殖阶段都达到峰值。Pearson相关性检验表明,雌鱼的雌二醇和睾酮与性腺成熟系数呈显著正相关关系(P<0.05);雄鱼的雌二醇和性腺成熟系数也存在显著正相关关系(P<0.05),而睾酮与性腺成熟系数和肥满度呈显著正相关关系(P<0.05)。3.克隆了Foxl2基因并分析其在大刺鳅繁殖周期中的表达变化。大刺鳅Foxl2基因全长1872 bp,包含一个918 bp的开放阅读框,编码305个氨基酸。大刺鳅Foxl2基因的C末端区域和叉头结构域是高度保守的,但是N末端区域与其它硬骨鱼类、鸟类、两栖动物和哺乳动物有较大差异。在不同组织中的qRT-PCR结果显示,在卵巢、鳃和眼中的表达量较高,且卵巢中的表达量最高,其次是脑和精巢,而在其它组织中的相对表达量较低。在早期发育过程中,Foxl2基因在原肠期的表达量显著升高,出膜后Foxl2基因的表达量开始增加,在25 d时表达量达到最高峰。大刺鳅Foxl2基因在卵巢成熟阶段的V期表达量最高,而精巢的表达模式就不同,在精巢发育阶段的III期表达量最高;在性腺成熟阶段,Foxl2基因在雌雄大脑中的表达量都较高。原位杂交实验结果显示,Foxl2基因主要在大刺鳅卵巢的滤泡细胞和精巢的间质细胞中表达。以上研究结果表明Foxl2基因在大刺鳅早期发育以及性腺发育和分化时期有着重要的作用。
