作者彭宏燕(2019)在《赤霉素处理条件下枇杷内源激素含量测定和Aux/IAA基因表达初步分析》一文中研究指出:三倍体枇杷由于配子母细胞减数分裂异常,难以形成正常的花粉,造成花粉败育而不能正常授粉受精,所以三倍体自然结实率极低,施用外源赤霉素(Gibberellins,GAs)后坐果率显著提高。已有研究表明,生长素(Auxin)是促进果实坐果和生长发育的重要内源激素之一,主要作用是促进细胞分裂。生长素/吲哚乙酸蛋白(auxin/indoleacetis acids proteins,Aux/IAAs)是生长素精确反馈调节的关键环节。为了从生理和分子水平探讨外源GA3诱导枇杷坐果过程中生长素信号转导途径的变化,本研究测定了GA3处理后子房壁厚度、内源吲哚-3-乙酸(indole-3-acetic acaid,IAA)含量以及生长素原初响应基因表达等指标的动态变化,旨在为三倍体无籽枇杷坐果机制研究提供理论参考。本试验以三倍体枇杷A322无性繁殖株系为研究对象,以清水处理为对照,分析了坐果期外源GA3处理后三倍体枇杷坐果率、子房壁厚度、子房和其他部分6种内源激素含量等形态学和生理指标的变化。同时,以前期转录组测序结果为依据,克隆了6个Aux/IAA家族基因的cDNA片段:IAA6,IAA8,IAA9,IAA10,IAA11和IAA20,进一步分析了这6个基因在三倍体枇杷坐果期的表达情况,主要研究结果如下:1.外源GA3处理后,三倍体枇杷坐果率显著提高,平均坐果率为43.32%,而对照组坐果率仅为1.9%。坐果期形态学观察发现,GA3处理7 d后萼片开始闭合,并在处理21 d后完全闭合。石蜡切片发现坐果期子房及周围组织细胞没有明显变化,通过测量坐果期子房壁厚度发现在坐果期子房壁未明显增厚,说明此时细胞分裂和膨大都很缓慢,处于幼果滞长期。通过测量坐果期胚珠纵横径发现处理组和对照组胚珠都没有继续发育。2.外源GA3处理后,子房内源IAA含量明显高于其它部分,并在处理后第3d和第5 d出现了两个高峰,推测其含量升高可能与外源GA3处理有关,高浓度的IAA替代受精作用从而启动枇杷坐果。对于脱落酸(Abscisic acid,ABA)含量变化而言,GA3处理后,子房中内源ABA含量整体较低;对照组(清水处理)在处理21 d后出现一个高峰,与落花时间一致。此外,枇杷坐果过程中玉米素(Zeatin,ZT)的含量无论是在子房还是在其他部分比另外五种激素高,推测内源ZT可能与枇杷坐果有关。3.基于转录组组装序列分析结果设计引物进行cDNA序列克隆,得到的IAA6与苹果MdIAA6(HM122447.1)相似性为93%;IAA8与苹果MdIAA8(NM001294015.1)相似性为97%;IAA9与苹果MdIAA9(NM001293892.1)相似性为93%;IAA10与苹果MdIAA10(HM122440.1)相似性为90%;IAA11与苹果MdIAA11(NM001293857.1)相似性为97%;IAA20与苹果MdIAA20(NM001293857)相似性为95%,其序列特征与预期结果一致。4.荧光定量PCR分析发现,GA3处理后6个基因的表达量都有明显变化,IAA6,IAA8,IAA10在子房中的表达量明显高于其它部分;IAA9在子房和其它部分的表达量相当,IAA11和IAA20在子房中的表达量低于其它部分。此外,在相同发育时期,外源GA3处理后子房中IAA9的表达量与内源IAA含量变化趋势一致,推测内源IAA含量升高,诱导IAA9表达上调,从而诱导枇杷果实坐果。子房中,IAA8在GA3处理3 d后表达量开始升高,并显著高于对照组,推测其功能可能与IAA9相似。
