作者王晓飘(2019)在《盐酸阿莫罗芬搽剂的研制》一文中研究指出:甲真菌病在临床上是一种常见的疾病,在皮肤真菌感染类疾病中大约占1/3,且在甲病变中也占到50%。患者感染甲真菌病后,生理上会产生负担,对心理上也有一定的打击。在生活中,致病菌也会为患者带来过敏反应,或者成为人群中的传染源。所以人们对甲真菌病的治疗也越来越迫切,成为了现在的重点研究项目。阿莫罗芬作为甲真菌病的局部治疗药物,对于轻度感染有着显著的疗效,能够广谱的杀菌或抑制细菌。目前临床上在不断研究各种新型的剂型,帮助药物更好的渗透进入甲板,达到治疗部位。除了单一治疗,还可以与其他口服药物联合治疗,如伊曲康唑、特比萘芬等。阿莫罗芬治疗的副作用较小,且对于浅部的真菌感染疗效更好,目前来看,也是治疗皮肤真菌一类疾病的理想药物,在临床市场上具有广阔的应用前景。盐酸阿莫罗芬搽剂因其特殊的辅料形成一种递药系统,涂抹在甲板上会形成一层薄膜,当溶剂蒸发后,药物可以长久的遗留在甲板上,渗透进入甲板,使药物作用在感染部位,增强药物的作用时间。本课题以盐酸阿莫罗芬为主药,以丙烯酸树脂RL100、三乙酸甘油酯、乙酸乙酯、乙酸丁酯以及乙醇为辅料研制出一种搽剂,并对其进行了以下研究:首先第一部分,对盐酸阿莫罗芬搽剂进行处方工艺的研究。根据甲板的特殊解剖结构,选择搽剂作为最佳剂型,易于涂抹,方便卫生。根据相关专利文献等确定初步处方,再以性状、pH值、相对密度等作为筛选指标,选择最优处方。第二部分,对盐酸阿莫罗芬搽剂的质量标准进行研究。参考原料药盐酸阿莫罗芬的质量标准,及剂型特点、中国药典搽剂的要求等,对盐酸阿莫罗芬搽剂各项目进行检测。本实验采用HPLC测定盐酸阿莫罗芬的有关物质和含量,对自制样品与市售样品进行检测,为盐酸阿莫罗芬搽剂的质量控制研究提供试验依据。第三部分,对盐酸阿莫罗芬搽剂进行稳定性试验的研究,从温度、湿度和光照三个条件下进行试验。稳定性的考察对药品的质量安全可控性奠定了坚实的基础。最后一个部分,对自制搽剂和市售搽剂进行了透甲试验的研究,并对比透甲量及透甲速率,对药物的吸收与渗透有参考性的意义。试验结果表明,自制搽剂与市售搽剂的渗透能力相似,可以认为二者在疗效上相当。
