作者何凌云(2019)在《大豆油或棕榈油替代鱼油对斜带石斑鱼生长性能、脂肪酸组成及脂肪代谢的影响》一文中研究指出:本试验以鱼粉、豆粕、酪蛋白和明胶为蛋白源,鱼油、豆油和棕榈油作为脂肪源,配制大豆油或棕榈油替代鱼油水平分别0、25%、50%、75%、100%9种等氮等脂试验饲料(对照组、25SO、50SO、75SO、100SO、25PO、50PO、75PO和100PO饲料组),将810尾初始均重约为12.59 g的斜带石斑鱼随机分到27个网箱中,每组3个重复,每个重复30尾鱼,每天早晚饲喂至表观饱食,试验周期为8周,比较探究大豆油、棕榈油替代鱼油对斜带石斑鱼生长性能、脂肪酸组成及脂肪代谢相关基因表达的影响及差异机制。主要研究结果如下:1.生长性能和体成分:无论大豆油还是棕榈油,随着替代鱼油水平增加,增重率(WGR)及肝脏与肌肉粗蛋白含量呈先升高后降低的趋势,但肝脏粗脂肪含量则呈逐渐升高趋势。5SO、100SO和100PO组肝脏粗脂肪含量显著高于对照组(P<0.05)。油源不影响斜带石斑鱼生长性能,但棕榈油组全鱼和肌肉粗蛋白含量高于大豆油组(P<0.05)。以增重率为测定指标,经过二次回归分析得到大豆油和棕榈油替代鱼油的最佳比例分别为32.9%和47.4%,以上结果表明,大豆油、棕榈油替代鱼油提高了肝脏粗脂肪含量,棕榈油组脂肪沉积高于大豆油组,棕榈油替代鱼油比例高于大豆油。2.血浆生化指标、肝脏抗氧化及脂肪代谢酶活:无论大豆油还是棕榈油,随替代鱼油水平增加,血浆T-CHO和HLD-C含量呈先升高后降低趋势,肝脏CAT活性和MDA含量呈逐渐降低趋势,而肝脏LPL和TL活性呈先降低后升高趋势。对照组和25SO组HLD-C、GSH-Px活性和MDA含量均显著高于100SO组(P<0.05),对照组HL活性显著低于100SO组(P<0.05),但50SO组中ACC和HSL活性显著低于100SO(P<0.05)。50PO组中血浆T-CHO和TG含量显著高于100PO组(P<0.05),50PO组TL活性显著低于75PO和100SO组(P<0.05),100PO组中血浆CAT活性和MDA含量显著低于对照组(P<0.05)。油源不影响肝脏代谢酶活性,但影响血浆TG含量和CAT活性和MDA含量。以上结果表明,25SO和50PO可相对提高机体脂肪代谢机能,100SO和100PO组的抗氧化能力和脂肪分解酶活性降低,脂肪酸合成酶活性升高。3.脂肪酸组成和肝脏组织形态:无论大豆油还是棕榈油,随替代鱼油水平的增加,肌肉和肝脏C18:1n-9和C18:2n-6含量呈逐渐升高的趋势,ARA、DHA、EPA含量呈逐渐降低的趋势。50SO-100SO组肌肉C18:1n-9含量显著低于对照组(P<0.05),50PO-100PO组肌肉ARA含量显著低于对照组(P<0.05);75PO和100PO组肝脏C18:1n-9含量显著高于对照组(P<0.05),50SO-100SO和100PO组肝脏C18:2n-6含量显著高于对照组(P<0.05),DHA均从50SO和50PO显著低于对照组(P<0.05)。油源明显影响肌肉中C18:1n-9、C18:2n-6、ARA、EPA、DHA含量以及肝脏C18:1n-9和C18:2n-6含量。肝脏组织结构中发现,与对照组比,100SO和100PO组出现细胞核偏移或消失,脂肪滴明显增加,细胞空泡化增加明显。以上结果表明,大豆油、棕榈油替代鱼油降低DHA和EPA含量,饲料脂肪酸含量对鱼体不同组织脂肪酸有不同的影响。4.肝脏代谢相关基因表达:随替代鱼油水平的增加,肝脏FAS、ACC、G6PD、PPARα、LPL、Δ6fad和SREBP-1c基因表达量均呈上升的趋势,但ATGL、FABP和HSL基因表达量呈降低的趋势,而CPT-1、Elovl5、Elovl4基因表达量呈现先升高后降低的趋势,SCD基因表达无变化。100SO组中FAS、ACC、G6PD和LPL基因表达量和100PO组FAS、ACC、G6PD、HSL、PPARα、LPL、Δ6fad和SREBP-1c基因表达量均显著高于对照组(P<0.05),但100PO和100SO组ATGL基因表达量和100PO组FABP和Elovl5基因表达量显著低于对照组(P<0.05),25SO组Elovl5和SREBP-1c基因表达量和25PO组Elovl4基因表达量显著高于对照组(P<0.05)。以上结果表明,100SO和100PO组上调了脂肪酸合成相关基因表达,下调了脂肪酸分解相关基因表达。综上,在本试验条件下,大豆油或棕榈油大比例甚至完全替代鱼油也不会对斜带石斑鱼生长产生明显影响,但降低了机体营养状况、抗氧化能力、脂肪分解酶活性和肝脏和肌肉DHA和EPA的含量,促进了肝脏脂肪沉积并造成肝脏组织结构损伤。
