作者张东栋(2019)在《以壳聚糖为基质的双层复合涂膜及电纺纳米纤维的研究》一文中研究指出:基于大分子聚合物制备的膜材料在保鲜领域的研究备受关注。本文分别采用层层自组装技术及静电纺丝技术制备基于壳聚糖的双层涂膜和电纺纤维膜,并添加溶菌酶,实现协同抑菌作用。以壳聚糖(CS)和海藻酸钠(ALG)两种带相反电荷的多糖为成膜基质,制备包埋溶菌酶(LZ)的ALG-CS+LZ双层复合涂膜,表征双层膜的微结构,评价膜的理化性能、抑菌性能及保鲜性能。以CS和聚乙烯醇(PVA)为基质,载入不同量LZ,制备系列LZ/CS/PVA电纺纳米纤维膜,分析纤维膜的形貌特点以及纤维膜的特性,为双层膜和电纺膜的进一步应用提供一定的理论基础。主要获得以下结果:1.采用层层自组装技术制备了ALG-CS+LZ双层复合涂膜,并对涂膜进行表征及理化性质的研究。结果表明:与CS、ALG、CS+LZ单层涂膜较光滑的表面相比,ALG-CS+LZ双层复合涂膜表面表现出明显的不规则分布和良好的立体效果,粗糙的表面有利于小分子物质的负载。双层复合涂膜横截面SEM图、FTIR谱图及XRD谱图表明ALG与CS+LZ涂层之间并非简单地叠加,而是ALG、CS、LZ分子之间存在相互作用力。双层涂膜的力学性能显著优于ALG涂膜,而接近CS+LZ涂膜;水蒸气透过率、氧气、二氧化碳透过率和透光率明显低于ALG涂膜,而接近CS+LZ涂膜。表明双层膜叠加理化性能并未降低。2.以水产优势腐败菌荧光假单胞菌和腐败希瓦氏菌为对象,研究双层复合涂膜的抑菌作用及机理。结果表明:与ALG,CS+LZ单层涂膜相比,ALG-CS+LZ双层复合涂膜对荧光假单胞菌和腐败希瓦氏菌的抑制作用更强,并且在作用初始12 h具有一定杀菌作用;双层涂膜导致细菌更多的核酸和电解质外漏,破坏细菌细胞膜的完整性,增加细胞膜通透性;并通过破坏或抑制维持细胞正常生命活动的蛋白质或酶及阻碍它们的合成等影响荧光假单胞菌、腐败希瓦氏菌生长。3.采用ALG、CS+LZ单层涂膜和ALG-CS+LZ双层复合涂膜处理大菱鲆鱼片,贮藏于4℃冰箱,每隔3天检测鱼肉中菌落总数(TVC)、pH值、挥发性盐基氮值(TVB-N)、硫代巴比妥酸(TBARS)、K值、质构、低场核磁共振分析等评测大菱鲆鱼片在冷藏期间的新鲜程度及质量的变化。结果表明:与ALG、CS+LZ单层涂膜液相比,ALG-CS+LZ双层复合涂膜更有效地抑制了大菱鲆鱼片中腐败微生物的繁殖;显著减缓了鱼片pH、TVB-N、K值等的增加;减少了鱼肉蛋白的变性和水分的散失,延长了大菱鲆鱼片的货架期4~6天。4.采用静电纺丝技术制备不同质量比的壳聚糖/聚乙烯醇纳米纤维膜(CS/PVA),在纤维形貌良好的配比溶液中添加不同含量LZ制备LZ/CS/PVA纳米纤维膜。采用SEM、FTIR、XRD及DSC等方法分析LZ/CS/PVA纤维膜形貌特点和纤维膜的特性。结果表明:LZ的添加影响了纤维形貌均匀程度及纤维之间的黏结程度;随着LZ的加入,纤维平均直径从339.62±92.72 nm(CS/PVA)降低至262.10±86.32~283.01±96.83 nm(LZ/CS/PVA系列纤维)之间;LZ添加量为0.30%和0.50%时,纤维形貌良好;纤维膜各组分之间存在强烈的相互作用,阻碍晶体的形成,致使其形成几乎无定形的结构;LZ的添加提高了纤维膜的热稳定性,熔点从185.60℃上升到189.30℃~191.00℃之间。LZ/CS/PVA纤维膜由安全无毒的材料制备而成,具有应用于食品工业的潜力。
