作者赖重波(2019)在《马齿苋对大鼠湿热泄泻模型的作用研究》一文中研究指出:本文以中草药马齿苋为研究主线,利用从仔猪泄泻肠道中及其他相关病料,分离、筛选出导致泄泻的的产ESBLs大肠杆菌构建大鼠湿热型泄泻模型,探讨马齿苋对该大肠杆菌构建的动物模型的治疗作用研究。试验主要方法和结果如下:1、从泄泻仔猪的病料中分离病原菌,经16sRNA鉴定为大肠杆菌。PCR检测其毒力基因,可知其含有细菌含有多重耐药调控基因marA、抗自由基基因sodA、卷曲菌毛基因csgA、脑膜炎相关和温度调节的菌毛基因mat、脂多糖(LPS)基因R1、外膜蛋白基因ompA、Ⅰ型菌毛(D-甘露糖特异性粘附)基因fimC、血清存活基因iss、耶尔森氏铁质基因fyuA这9个毒力因子。该大肠杆菌的致泻毒力可能与R1、csgA、fimC有关。2、采用二倍微量稀释法对受试大肠杆菌进行抗菌药、马齿苋水提物MIC的检测;再以马齿苋水提物的1/2MIC、抗菌药的2MIC进行细菌传代试验。对该菌株的耐药基因进行PCR扩增检测。结果得该大肠杆菌携带耐药基因有:磺胺类:sul2;四环素类:tetA;β-内酰胺类:CTX-M-U;质粒介导喹诺酮类:qnrD、qepA;耐磷霉素:fosA3;是一株多重耐药的产ESBLs大肠杆菌。根据2018版CLSI标准1,受试菌株对以上类别抗菌药均表现为耐药,而传代试验则表明,马齿苋水提物能增强受试菌株对大部分抗菌药的敏感性,使其MIC下降。3、动物模型试验:将SD大鼠随机的分为6组:Ⅰ空白对照组,Ⅱ模型组,Ⅲ自愈组,Ⅳ马齿苋低剂量治疗组,V马齿苋中剂量治疗组,Ⅵ马齿苋高剂量治疗组。通过对大鼠实行高脂饮食、高温高湿环境和腹腔注射致泻型大肠杆菌液构建湿热型泄泻大鼠模型;按照低、中、高剂量,用马齿苋的水提取物对试验大鼠进行灌胃治疗。对各组大鼠血常规、肝肾功能指标、炎症因子(TNF-α IL-6、IL-17、IL-23)、脏器系数(肝、脾、胸腺)以及各脏器的病理组织学进行检查。结果显示:模型组符合湿热型泄泻标准,判定模型建立成功;试验大鼠机体出现了全身性炎症、血脂水平上升,免疫功能下降,病理组织学检查可见多脏器出血、变性、坏死。马齿苋水提物通过促炎症因子的抑制,对大鼠炎症网络功能的调控,对免疫器官的恢复保护功能,并具有止泻、降血脂、降体温的功效;对脏器的出血、变性情况也有一定的改善作用;并且马齿苋的疗效随着剂量的增加而增强。表明马齿苋对湿热型泄泻模型具有良好的治疗作用。本试验初步探究了马齿苋中药对产ESBLs大肠杆菌构建的大鼠湿热型泄泻模型的作用,为马齿苋中药的对大肠杆菌体内应用提供参考。
