谭志承:限食对家蚕血淋巴代谢生理和个体寿命的影响论文

作者谭志承(2019)在《限食对家蚕血淋巴代谢生理和个体寿命的影响》一文中研究指出:限制热量,减少食物摄入已在多种动物模型中被证明能够有效延缓衰老和延长寿命[1]。但是限食延长寿命的机理,至今仍未明确,还有许多未知环节需要继续深入研究。家蚕作为一种饲养容易、子代多、遗传背景清楚和突变资源丰富的实验动物,用来研究延缓衰老和延长寿命具有自己的独特优势。家蚕二化性品系“秋丰”幼虫具有限性斑纹,雌性为普通斑纹,雄性为素蚕,便于区分雌雄。本文以家蚕品种秋丰为实验对象,利用qRT-PCR和代谢组分析技术,研究限食对家蚕幼虫血淋巴代谢生理、寿命相关基因表达水平的影响,探究限食与寿命相关基因的关联作用,寻求限食延缓衰老的生理生化机制。主要研究内容和结果如下:(1)限食显著减轻家蚕个体质量、显著延长家蚕寿命。以二化性家蚕品系“秋丰”为对象,通过前期预备试验,设置自由采食组(AL组)和限制给食组(DR组),AL组每日给桑2次、保持24 h自由进食;限制给食组(DR组)每日给桑1次、保持16h自由进食时间即停食8 h。结果显示,DR组给食量控制在AL组给食量的61.03%;与AL组相比,DR组5龄幼虫个体质量平均减轻36.41%,雌雄平均寿命955 h,比AL延长131 h,差异达到显著水平(p<0.05);2组雌蛾的造卵数没有明显变化(p>0.05),雄蛾交尾正常。(2)限食影响家蚕3个寿命相关基因的表达。选择3个果蝇寿命相关基因的同源基因Bmchico、Bmrpd3和BmAkh2。分别提取DR组和AL组5龄3 d幼虫不同组织的总RNA,利用qRT-PCR技术分析其表达水平。结果显示,Bmchico在DR组雌蚕丝腺、脂肪体和雄蚕表皮中显著下调,在其他组织中下调不明显;Bmrpd3在2组家蚕的各个组织中观察到不同程度的表达差异;BmAkh2在除DR组雄蚕表皮的其余组织中显示出不同程度的下调。表明限食对3个寿命相关基因在不同性别、不同组织的蚕体存在不同影响。(3)限食对家蚕生理生化水平的影响。分别提取DR组和AL组5龄3 d幼虫血淋巴,测定超氧化物歧化酶活力、过氧化氢酶活力,丙二醛含量、可溶性蛋白含量和海藻糖水平。结果显示,DR组血淋巴中超氧化物歧化酶和过氧化氢酶活力整体水平较AL组高,丙二醛含量下降,可溶性蛋白含量降低,海藻糖整体水平下降。表明限食后生物体具有更高的抗氧化能力,更低的能量代谢水平。(4)限食对家蚕血淋巴代谢物的影响。本实验选取DR组和AL组幼虫5龄3 d血淋巴进行LC-MS代谢组学分析。结果显示,在雄蚕中筛选到了74种差异代谢物(26种上调,48种下调),在雌蚕中筛选到61种差异代谢物(22种上调,39种下调),其中主要的差异代谢物是氨基酸、有机酸和胺类物质。在DR组中,组氨酸、精氨酸等17种氨基酸含量发生显著改变,各类氨基酸所占比重重排;柠檬酸、苹果酸、α-酮戊二酸等TCA中间体含量上升;精胺、腐胺和亚精胺等生物胺整体水平下降。研究表明限食可以提高家蚕机体抗氧化水平,改变血糖水平,使家蚕血淋巴代谢物产生显著变化,从而延长家蚕的寿命。该结果为人类限食研究提供了良好的动物模型,为研究适度限食有助于抗衰老提供了实验依据,为限食延缓衰老机制理论基础的后续研究打下基础。

Abstract
