作者毛立霞(2019)在《大白菜根肿病抗性基因的精细定位与标记开发》一文中研究指出:根肿病是由根肿菌侵染芸薹属植物的根部引起的,被称为芸薹属植物的“癌症”。大白菜属于芸薹属植物,产量受根肿病的影响十分严重。近年来,大白菜根肿病发病面积急剧增加,造成了重大的经济损失。因此,抗性基因的挖掘和抗根肿病品种的培育在大白菜根肿病的研究中具有十分重要的意义。本研究以抗根肿病材料欧洲芜菁ECD01和感病DH系Y510-9作为研究亲本,构建F2群体以及F2:3家系,利用BSA-Seq对抗根肿病基因Crr5进行定位,开发与之紧密连锁的分子标记,用于大白菜分子辅助育种。主要得到以下结果:1.对感病材料YX55、Y510-9和抗病材料ECD01、14CR514M-3被根肿菌侵染的根毛及皮层进行细胞学鉴定。结果表明感病材料YX55的平均根毛侵染率比抗病材料ECD01、14CR514M-3的平均根毛侵染率高47.53%-50.02%,Y510-9的平均根毛侵染率比ECD01的高40.06%。从皮层侵染来看,从第6天就观察到感病材料YX55、Y510-9的皮层被侵染。2.对F2群体进行根肿病鉴定和抗性遗传分析,经卡方检验,分离比例符合3:1,表明该根肿病抗性基因是由单基因控制的。构建极端DNA混池,进行BSA-Seq分析,将根肿病抗性基因定位于A08染色体的0.4-2.2 Mb之间,并将该基因命名为Crr5。3.开发20对多态性标记及用课题组已有的大白菜A08染色体上的标记筛选了5对多态性标记,用JoinMap 4.0对标记进行连锁分析,将Crr5基因定位在标记KMM32-4和Bra154-4之间,遗传距离分别是0.1 cM和0.4 cM,物理位置在1.1-1.6 Mb。利用开发的最佳标记之一KMM32-4对8个BC2F2群体进行正确性验证,其一致率在91.11%-100%之间,平均一致率为96.78%,该标记初步用于分子标记辅助选择。4.在1.1-1.6 Mb有57个预测基因,其中BraA08000154基因编码产物包含Leucine-rich repeat结构域,可能参与抗病过程。对这个基因的相对表达量进行分析。结果表明,同一时期这个基因在抗病植株的表达量显著高于感病植株的表达量,进一步表明这个基因可能为根肿病抗病基因Crr5的候选基因。
