作者何艳(2019)在《多花黄精组织培养体系的研究》一文中研究指出:多花黄精(Polygonatum cyrtonema)为百合科(Liliaceae)黄精属(Polygonatum)多年生草本植物。其根状茎肥厚,是2015年版《中国药典》中药黄精的三种基原植物之一。在大健康背景下,黄精产业已经进入了飞速发展的新时期,但传统的种子或块茎繁殖方式存在资源浪费、种性退化等问题。关于多花黄精的组织培养技术已有报道,但均以其块茎为外植体,且存在污染控制难、繁殖系数低、生产周期长等难点,难以满足种苗工业化生产需求,导致优质种苗十分缺乏。本研究以多花黄精块茎为外植体,优化其组织培养体系,在此基础上开辟以无菌苗叶片为外植体的新快繁体系,并对其愈伤组织形成和分化过程进行了细胞学研究,为多花黄精优质种苗生产提供了技术支撑和遗传转化基础。主要研究结果如下:(1)通过块茎外植体灭菌优化试验发现,未经1‰多菌灵溶液浸泡的块茎污染率基本为100%,经过1‰多菌灵溶液浸泡48h后,再用0.15%氯化汞(10min)和2.5%次氯酸钠(5min)复合消毒,污染率显著下降,仅为29.33%;(2)对块茎不定芽分化培养基进行筛选,得到不定芽分化最佳培养基为MS+6-BA 1.0 mg·L-1+NAA 1.0 mg·L-1+TDZ 1.0 mg·L-1,分化率为88.67%,平均芽数3.58个,且芽生长状况良好;(3)以多花黄精无菌苗叶片为外植体的组织培养研究发现,30d叶龄叶基在MS+6-BA 1.5 mg·L-1+2,4-D 0.2 mg·L-1培养基上愈伤组织诱导率最高,达到80%;叶片不定芽最佳分化培养基为MS+6-BA 4.0 mg·L-1+2,4-D 0.2 mg·L-1上,分化率为90%,平均分化芽数5.16个;不定芽最佳增殖培养基为MS+6-BA 2.0 mg·L-1+NAA 0.10.2mg·L-1;切分三叶无菌苗在培养基1/2MS+IBA 2.0 mg·L-1中,生根率最高,为94%,平均根数8.3条,平均根长4.07cm;生根无菌苗移栽于泥炭-蛭石1:1的基质中生长良好,40d后存活率达85%以上;(4)对多花黄精无菌苗叶片离体培养过程的细胞学观察发现,30d叶龄叶片基部的细胞具有分生能力强的特点,能产生愈伤组织;不定芽有外起源和内起源两种发生方式,分别由愈伤组织近表层细胞和愈伤组织内部拟分生组织形成,且外起源发生方式早于内起源。
Abstract
