论文摘要
目的:探讨丹参素对转化生长因子β1(TGFβ1)刺激大鼠肝星状细胞(HSC)信号传导通路的影响。方法:体外分离、培养大鼠肝HSC,以MTT法检测丹参素对HSC增殖的影响及丹参素对TGFβ1刺激HSC增殖的影响;用免疫细胞化学染色法观察丹参素对TGFβ1刺激HSC表达α-SMA的影响;以RT-PCR法分析丹参素和TGFβ1作用HSC后,其Smad2、Smad3、Smad7 mRNA表达的变化。结果:(1)丹参素在0.0625 mmol/ L~1mmol/ L时,对HSC的生长增殖具有抑制作用,其作用强度随丹参素浓度增加增强(P<0.05);丹参素对TGFβ1引起的HSC增殖也具有明显的抑制作用,并呈剂量依赖性(P<0.05)。(2)丹参素0.25 mmol/ L作用HSC能下调α-SMA的表达,也能下调TGFβ1诱导的HSC的α-SMA表达(P<0.05),从而抑制HSC活化。(3) TGFβ1促进HSC中Smad2、3、7 mRNA的表达;丹参素能下调TGFβ1诱导的HSC内Smad2、Smad3 mRNA的表达(P<0.05),并能上调Smad7 mRNA表达(P<0.05).结论:丹参素能直接或间接地上调HSC内Smad7 mRNA表达,并下调Smad2、Smad3mRNA表达,恢复正常的TGFβ1跨膜信号转导,抑制HSC活化和TGFβ1促进HSC活化的作用。
