作者贺栾劲芝(2019)在《黄瓜果实变异突变体的筛选》一文中研究指出:黄瓜(Cucumis sativus L.,2n=2x=14)是世界十大蔬菜之一,也是我国保护地栽培的主要蔬菜作物。2017年我国黄瓜栽培面积为123.7万公顷,年产量达6487.5万吨,占世界总量的54.4%和77.5%。黄瓜食用部分是幼嫩的果实部分,果实性状是重要的农艺性状,直接影响经济效益。利用EMS诱变技术创制新种质,构建黄瓜突变体库,从中筛选果实变异突变体,为果实发育机理研究及黄瓜育种提供优异材料。本文以前期构建的黄瓜自交系‘6457’突变体库M2代群体为材料,连续两代自交纯化,对盛果期商品瓜及种子成熟期种瓜进行表型鉴定和数据调查,主要结果如下:1.在日光温室栽培条件下,以M2代群体为材料,经过一次自交,构建包含176个变异株系的M3代群体,从中筛选出果实性状发生变异的典型株系44个,构成黄瓜果实变异突变体库M4代群体。2.在日光温室栽培条件下,以构建的528株黄瓜果实变异突变体库M4代群体为材料,以野生型‘6457’为对照,在盛果期对果实性状进行鉴定,(1)瓜把形状变异1种(溜肩形),瓜色变异2种(白绿色、绿色),瓜刺瘤稀密变异3种(无刺、稀刺、密刺),瓜刺变异1种(软毛刺),瓜瘤变异3种(无瘤、中瘤、大瘤)等共13种典型果实变异;(2)纯合株系26个,占59.1%,从中筛选出果实短圆筒形材料1份,果实纺锤形1份,果实无刺瘤材料2份,软毛刺材料1份等特异新种质;(3)野生型平均瓜长为23.0 cm,突变体库瓜长分布为7.525.7 cm,其中突变体m18s55瓜长最短(9.0 cm),仅为野生型果实长度的39.1%;(4)野生型平均瓜横径为3.0 cm,突变体瓜横径分布1.84.3 cm,其中突变体m18s55瓜横径最大(5.1 cm),为野生型的1.7倍,突变体m18s9的瓜横径最小(1.8cm),为m18s55瓜横径的35.3%。3.对种子成熟期种瓜瓜长、瓜横径、果肉厚度、瓜把、果柄等进行数据调查与分析,筛选出果实变长材料2份,果实变短材料5份,瓜横径变大材料1份,瓜横径变小材料5份,瓜肉厚变小材料3份,瓜把性状相关材料14份,果柄变长材料1份,果柄变短材料1份。(1)野生型平均瓜长为29.2 cm,突变体库瓜长分布为10.038.1 cm,其中突变体m18s18果实长度最长(35.5 cm),突变体m18s55瓜长最短(10.0 cm),仅为最长瓜的五分之一;(2)野生型平均瓜横径为5.3 cm,突变体库瓜横径分布为4.07.5 cm,其中突变体m18s56瓜横径最大(7.3cm)。突变体m18s55瓜横径最小(4.0 cm)。(3)野生型平均瓜肉厚为0.9 cm,突变体库瓜肉厚分布为0.51.5 cm,其中突变体m18s55瓜肉厚最小(0.5 cm)。(4)野生型平均瓜把长为4.4 cm。突变体库瓜把长分布为2.05.3 cm。其中突变体m18s61瓜把长最长(6.0 cm),为野生型的1.4倍,突变体m18s22、m18s55无瓜把;野生型平均瓜把宽度为3.0 cm。突变体库瓜把宽度分布为2.75.7 cm。其中突变体m18s36瓜把宽度最小(2.6 cm)。(5)野生型平均果柄长为3.1 cm。突变体库果柄长分布为0.84.5 cm。其中突变体m18s39果柄长最长(3.9 cm),为野生型的1.3倍,突变体m18s55果柄长度最短(0.8 cm),仅为果柄最长突变体的五分之一。
Abstract
