作者李洪林(2019)在《贵州地方鸡种隐性白羽与慢羽分子标记研究与应用》一文中研究指出:为了准确鉴定兴义矮脚鸡白羽系的白羽性状,本文根据TYR基因序列和PMEL17序列各分别设计了特异性引物各1对,采用PCR直接测序法鉴定兴义矮脚鸡白羽系(以下简称XB)和罗曼蛋鸡(以下简称LB)的羽色控制基因,建立了地方鸡种显性白羽和隐性白羽分子鉴定方法,开展了罗曼蛋鸡与兴义矮脚白羽系杂交后代中隐性白羽纯合子(以下简称YXB)的鉴定和挑选工作。为了鉴定贵州黄鸡慢羽系、瑶山鸡慢羽系的慢羽性状,根据控制慢羽性状的基因序列,设计特异性引物1对,采用PCR直接测序法对表型鉴别的贵州黄鸡慢羽系(以下简称GHS)和瑶山鸡慢羽系(以下简称YSS)个体进行了分子鉴定,同时设计引物鉴定了慢羽公鸡纯合子,建立了两个鸡种慢羽性状分子标记鉴定方法;统计了贵州黄鸡、瑶山鸡不同世代慢羽羽型的变化规律,探讨了不同羽型对羽毛和体重的影响。结果如下:(1)经分子鉴定,检测到兴义矮脚鸡白羽系中TYR基因196bp的插入片段,测序后比对与原序列一致,未检测到PMEL17基因的160bp插入片段,判定兴义矮脚鸡白羽系为TYR基因控制的隐性白羽鸡,基因型为iicc;检测到罗曼蛋鸡PMEL17基因160bp的插入片段,测序后比对与原序列一致,判定罗曼蛋鸡为PMEL17基因控制的显性白羽,基因型为IICC;对罗曼蛋鸡和兴义矮脚鸡白羽系杂交2代208只鸡的TYR基因进行检测,发现存在IiCC、IICC等8种基因型,未检测到iiCC型。最终鉴定出iicc型个体9只,占总检测数的4.3%,经X~2检验,理论值与实际值相符(P>0.05)。(2)根据控制慢羽性状的基因序列,设计特异性引物1对,经PCR扩增在贵州黄鸡、瑶山鸡慢羽群体中发现1305bp的插入片段,测序比对后与原序列一致,此片段可作为慢羽性状检测的标记片段。以此片段为依据,对表型鉴定的贵州黄鸡慢羽和瑶山鸡慢羽进行分子鉴定,发现贵州黄鸡慢羽系表型鉴定的准确率为98.3%,瑶山鸡慢羽表型鉴定的准确率为97.9%。(3)为了验证慢羽个体分子鉴定的准确性,选择贵州黄鸡8只快羽公鸡,48只慢羽母鸡进行杂交,对146只杂交后代快慢羽性状进行表型鉴定,发现慢羽公鸡67只,快羽母鸡77只,X~2检验,理论值与实际值相符(P>0.05);选择瑶山鸡6只快羽公鸡,36只慢羽母鸡进行杂交,对131只杂交后代快慢羽性状进行表型鉴定,发现慢羽公鸡65只,快羽母鸡66只,X~2检验,理论值与实际值相符(P>0.05)。(4)研究发现,贵州黄鸡、瑶山鸡慢羽羽型在不同世代间变化较大,贵州黄鸡、瑶山鸡慢羽系原代以等长型和倒长型为主导羽型,分别占总慢羽羽型的94.07%和93.67%;经分子标记鉴定后,一世代以倒长型和未长出型为主导羽型,分别占总慢羽羽型数的95.07%和94.98%。为了研究不同羽型对羽毛生长和体重的影响,试验统计了贵州黄鸡0-49日龄羽毛生长和体重发育情况。在主翼羽中发现,28日龄前(除14日龄外),快羽型的主翼羽生长速度明显快于慢羽型的主翼羽(P>0.05);在尾羽中发现,21到42日龄之间,快羽羽型的主尾羽明显长于慢羽其他羽型(P<0.05),而49日龄时,未长出型个体覆主翼羽明显长于倒长型个体(P<0.05);慢羽羽型对体重影响不大,各羽型间差异不显著(P>0.05)。本文建立了隐性白羽和慢羽性状鉴定的分子标记,经验证准确,可用于生产实践。
