作者谢恩俊(2019)在《中黄1号茶树组培快繁及再生体系建立》一文中研究指出:中黄1号茶树是山茶科山茶属多年生常绿灌木,氨基酸含量高,茶多酚和咖啡碱含量适中,抗逆性强,是我国选育的特色珍稀黄茶品种。传统生产上采用扦插的繁殖方式繁殖率较低,不能满足茶苗的需求。本研究以中黄1号茶树为材料,探索其组织培养的基本条件,建立了中黄1号茶树茎段为外植体的组培快繁技术体系,为中黄1号茶树种质的生产提供技术支持,研究同时建立中黄1号茶树胚轴再生体系,为茶树遗传转化奠定基础。主要研究结果如下:(1)中黄1号茶树茎段组培快繁体系建立以中黄1号茎段为外植体,探索外植体的取样时间、消毒条件、植物生长调节剂对腋芽萌发、增殖、壮苗和生根的影响。结果表明,在4、6、8月份采取的幼嫩茎段污染率最低的为4月份采取的外植体。茎段组培无菌体系探索表明,茎段经75%酒精浸泡60 s,0.1%升汞浸泡12 min,20 d后外植体存活率为91.3%,污染率为2.7%。对腋芽萌发及增殖培养最佳培养基探索表明,消毒的无菌茎段接种于腋芽萌发培养基MS+2mg.L-16-BA+1mg.L-1GA3,培养40 d出芽率为91.7%,待腋芽长至2-4 cm后,从茎段上切下接种于增殖培养基诱导丛生芽,本研究条件下的最适腋芽增殖培养基配方为MS+4mg.L-16-BA+1mg.L-1IBA,培养45 d增值系数3.8,60 d后丛生芽长至2-4 cm,将芽分开进行壮苗和生根培养,见(3)。由此得出,中黄1号组培快繁体系建立提高了茶苗繁殖率,缩短育苗周期。(2)中黄1号茶树胚轴组培再生体系建立以中黄1号茶树种子为外植体,探索外植体的消毒条件和处理方式、植物生长调节剂对种胚萌发、胚轴愈伤组织诱导与分化、壮苗和生根的影响。结果表明,子叶和种胚一起经75%酒精浸泡1.5 min,0.1%升汞浸泡20 min,去除子叶的种胚,污染率低且存活率较高,20 d后外植体存活率为90%,污染率为6.7%。对种胚萌发条件探索表明,无菌种胚接种于萌发培养基MS+1mg.L-1GA3,培养15 d,萌发率可达98.8%,种胚萌发整齐,胚轴较长,60 d后,以萌发培养基中生长无菌茶苗的胚轴为外植体,对胚轴愈伤组织诱导及分化条件探索表明,胚轴愈伤组织诱导及分化最适培养基配方为MS+2mg.L-16-BA+0.3mg.L-1IBA,此时愈伤紧实、颜色翠绿,不定芽长势良好,出愈率为95.5%,分化率为17.5%,待不定芽长至2-3 cm将芽分开进行壮苗和生根培养,见(3)。(3)中黄1号组培苗壮苗与生根培养以中黄1号茶树茎段诱导的丛生芽和胚轴愈伤组织分化获得的不定芽进行壮苗培养,研究6-BA和IBA两种不同浓度配比的植物生长调节剂对组培苗生长以及生根的影响。结果表明,最适壮苗培养基配方为MS+1mg.L-16-BA+0.5mg.L-1IBA,培养45 d芽苗平均高度为5.3 cm,苗健壮。对组培苗诱导生根条件探索表明,植株在50mg.L-1IBA浸泡5 min,后转至1/2MS+1.5mg.L-1IBA培养基中,诱导生根率达到82.5%,平均生根5-6条,主根长,侧根较多。由此得出,该激素浓度条件可以很好的促进中黄1号组培苗生长和根的分化。(4)中黄1号组培苗炼苗和移栽以中黄1号组培苗为材料,研究不同炼苗时间和移栽基质对组培苗成活率的影响。结果显示,自然光条件下开瓶室温炼苗5 d,移栽到以营养土和蛭石2:1比例基质中,移栽成活率达66.7%,说明该条件下提高茶苗移栽成活率。
