辛伐他汀对高血压大鼠主动脉的保护机制

目的:研究他汀类药物对核因子κB(NF-κB)、单核细胞趋化蛋白-1(MCP-1)在自发性高血压大鼠(SHR)大血管中表达的影响,探讨其预防高血压所伴发的动脉粥样硬化的可能机制。 方法:20只12周龄雄性SHR被随机分为SHR组和辛伐他汀(simvastatin)组。另取10只同龄雄性京都种Wistar大鼠为WKY组。simvastatin组用simvastatin 5mg/kg·d灌胃共8周,SHR组和WKY组灌等量生理盐水作对照共8周,免疫组织化学法检测各组大鼠主动脉组织中NF-κB、MCP-1的表达,并用ELISA法检测各组血清中MCP-1的含量。 结果:与WKY组比较,SHR组主动脉组织中NF-κB、MCP-1表达及血清MCP-1含量显著增加(P<0.01),且组织中MCP-1的表达与NF-κB活化呈显著正相关(r=0.728,P<0.01)、血清MCP-1含量与其组织中MCP-1阳性信号指数也呈正相关(r=0.683,P<0.05):与SHR组比较,simvastatin组主动脉组织中NF-κB、MCP-1表达和血清MCP-1含量显著降低(P<0.01)。WKY组、SHR组及simvastatin组之间血清总胆固醇、总甘油三脂水平无明显差异(P>0.05)。SHR组及simvastatin组之间血压水平无明显差异(P>0.05)。 结论:自发性高血压大鼠主动脉组织中NF-κB、MCP-1表达增加,更为重要的是,3-羟-3-甲基戊二酰辅酶A还原酶抑制剂辛伐他汀可以明显抑制高血压所致的NF-κB的活化和MCP-1表达,且不依赖于高血压大鼠的血压及血脂水平。他汀类药物可能独立于降脂外通过抑制NF-κB的活化来调控MCP-1表达而起到大血管保护作用。
