作者惠建超(2019)在《高活性核桃内生真菌筛选及其代谢产物抑菌机理研究(Ⅱ)》一文中研究指出:核桃内生真菌具有丰富的生物多样性,为了更好地利用这种微生物资源,本研究在核桃内生真菌活性菌株筛选、发酵条件优化、代谢产物分离纯化,以及抑菌机理等方面进行研究,主要结果如下:(1)采用生长速率法,以6种常见植物病原菌为指示菌,对16株核桃内生真菌进行活性筛选。结果表明,1株来源于良种核桃树根部的内生真菌G3,其发酵产物对6种病原菌均表现出一定的抑菌活性。其中对苹果腐烂病原菌抑制率达到100%,完全抑制了病原菌菌丝生长;对水稻纹枯、核桃炭疽病原菌抑制率均大于75%。通过形态学特征观察结合分子生物学鉴定,G3确定为弯角镰刀菌Fusarium camptoceras。(2)以接种量、发酵温度和发酵天数为影响因子,以G3发酵产物对苹果腐烂病原菌的抑制率为评价指标,通过单因素试验、正交试验对核桃内生真菌G3的发酵工艺条件进行优化。结果表明,G3发酵的最优工艺为:接种量10%,发酵温度28℃,发酵时间30 d;此工艺条件下,G3发酵产物对苹果腐烂病原菌抑制率为85.17%。影响因子对抑制率的影响依次为:发酵温度>发酵天数>接种量。(3)采用液液分配方法,对核桃内生真菌G3固体发酵产物的乙醇提取物依次用石油醚、乙酸乙酯、正丁醇分别进行萃取。结果表明,正丁醇相和乙酸乙酯相对苹果腐烂病原菌抑制的IC50值分别为3.3 mg/mL和4.3 mg/mL;结合各萃取相质量及其对苹果腐烂病原菌抑制效果综合考虑,选择正丁醇相和乙酸乙酯相作为深入研究的活性萃取相。(4)通过对各萃取相化学成分预试验,结果显示,正丁醇相主要含有有机酸类物质,乙酸乙酯相主要为酚类物质,推测G3发酵产物活性物质可能为有机酸与酚类物质。对乙酸乙酯萃取相进行柱色谱分离获得高活性组分Fry2-2,供试浓度为1 mg/mL时,对苹果腐烂病原菌抑制率达到89.36%;GC-MS分析共检测到15种化合物,主要为胺类、脂类、酚类与烷烃类。对Fry2-2进一步分离,在Fry2-2-2组分中析出化合物A,经过重结晶纯化和核磁共振等检测,化合物A鉴定为:2,2’-亚甲基双[6-(1,1-二甲基乙基)]-4-甲基-苯酚,抑制苹果腐烂病原菌的IC50值为0.11 mg/mL。(5)在体外和体内条件下,对G3高活性组分抑制苹果腐烂病的机理进行研究。结果表明:高活性组分处理下,病原菌菌丝出现破损、撕裂,或者顶端形成纺锤形,使其生长异常,菌丝量减少;病原菌菌液的电导率增加、麦角甾醇含量降低、几丁质酶活性增加;感病苹果的体内防御酶SOD、PPO、POD活性均明显升高,高活性组分与5 mmol/L水杨酸两者处理效果相近,防御酶活性都呈现先升高后降低的趋势,并能够明显抑制苹果腐烂病斑的扩展,最大抑制率为77.61%。
