针对远交林木群体构建连锁不平衡图谱

随着分子标记的产生,运用分子标记进行植物QTL的研究取得了巨大进展。对林木的分子遗传研究和用于分析林木分子标记数据的统计模型尽管落后于农作物,但因其多数都是野生或半野生状态,这就为QTL定位提供了丰富的自然变异,许多统计方法随之应运而生,其中,连锁不平衡作图方法在近来定位复杂性状的基因组研究中已经成为一个主要焦点。本研究就是在连锁不平衡作图的理论基础上,通过一个自由授粉林木子代的遗传设计,提出两个统计模型用于野生种群(包括林木)的子代检验,进而可用于QTL的检测、定位及其效应分析。我们采用最大似然方法、EM算法以及假设检验对标记之间的连锁、连锁相和连锁不平衡同时进行估计,从而得到重组率和异交率等遗传参数的最大似然估计值,并将两点分析模型扩展到三个位点的分析,通过蒙特卡罗模拟方法模拟不同样本量和异交率情况下的遗传参数的估计情况,进而证实模型的统计性质,通过模拟比较,发现三点分析更为精确,这一模型及其扩展可以很好的用于以后的QTL检验和定位。我们用此模型来分析裸子植物香榧,构建了相应的遗传连锁图谱。本研究提出的模型和算法能够很好的应用于林木的QTL定位上,并且自由授粉子代的设计和一系列分析检验策略为定位控制植物复杂性状的QTLs提供了标准的程序。
