作者赵晓晓(2019)在《柳枝稷幼穗分化分期与TIFY基因家族鉴定》一文中研究指出:柳枝稷(Panicum virgatum L.)为禾本科多年生C4草本植物,近年来国际上已将其作为模式能源作物进行深入研究,但对柳枝稷生殖生长特性的研究相对较少。幼穗分化是营养生长与生殖生长的转折点,正确判断幼穗分化关键期对作物的生长发育调控、栽培管理与育种研究意义重大,可目前国内外尚未开展柳枝稷幼穗分化过程分期的研究。生殖生长是个体发育的重要阶段,受外界坏境和内部因素的共同调控,TIFY基因家族作为转录因子或转录因子互作蛋白,参与调控植物的生长发育、激素应答及胁迫响应等过程,然而柳枝稷TIFY基因家族研究尚未见报道。本研究为探索柳枝稷幼穗分化规律及TIFY基因家族在柳枝稷生殖生长过程中的功能,根据柳枝稷幼穗分化各阶段组织形态显微观察结果将柳枝稷幼穗分化过程进行了分期,并记录了对应的大田植株形态以供判别分化进程;对柳枝稷基因组中的TIFY基因家族成员进行了鉴定,并分析了各成员的进化关系、结构组成及表达特性;选定水稻中两个与花序发育有关的JAZ亚家族成员进行了同源克隆,并对两者的分子特征及亚细胞定位进行了分析且构建了基因过表达载体。论文从形态观察与分子调控两方面对柳枝稷的花序发育进行了研究,为柳枝稷生殖生长特性的探索奠定基础。主要结果如下:1.柳枝稷幼穗分化是一个慢-快-慢的过程,可分为9个连续的时期:苞原基分化期、枝梗原基分化期、小穗分化期、小花原基分化期、雌雄蕊原基分化期、药隔形成期、花药四分期、花药伸长期、羽状柱头形成期;每个时期均具有与之对应的植株外观形态以供判别幼穗分化进程:苞原基分化期至药隔形成期,叶龄、叶龄余数及可见叶数可用来判断幼穗分化所处时期,花药四分期至羽状柱头形成期,花序与小穗的长度及形态可用来辨别幼穗分化进程;柳枝稷幼穗分化起始至分化完全历时约3个月,植株历经拔节、孕穗、抽穗而开花,花序从上到下依次开花,花期可达3-4周。2.柳枝稷中共有48个TIFY成员,分属于TIFY(1个)、ZML(7个)、JAZ(40个)3个亚家族;PviTIFY基因分布在除染色体3以外的各条染色体上,其中半数成员集中定位在染色体9;PviTIFY蛋白依据进化关系及结构特点可分为5组:JAZⅠ、JAZⅡ、JAZⅢ、TIFY、ZML,每组内亲缘关系较近的成员具有相似的基因结构及保守基序组成;PviTIFY启动子区存在多种胁迫响应元件与激素信号调控元件,经定量实验证实PviTIFY10成员在JA处理及盐与干旱胁迫下反应强烈;JAZ亚家族部分成员尤其是PviTIFY10b,在花序与根的发育过程中有较高表达。根据PviTIFY的表达特性推测其可能在柳枝稷激素信号调控、胁迫防御反应及生长发育过程等方面发挥功能。3.成功克隆OsJAZ1与OsTIFY10b的同源基因PviTIFY3a与PviTIFY10b;两个基因所编码蛋白的保守结构域组成、进化特点、理化性质及二级结构组成等分子特征相似;PviTIFY3a-GFP融合蛋白瞬时表达结果显示PviTIFY3a定位于细胞核与细胞膜,软件预测PviTIFY10b很可能定位于细胞质;两个基因的过表达载体均已成功构建。
