作者华一杉(2019)在《TLR接头分子的起源进化及日本七鳃鳗TICAM-a和TICAM-b的功能初探》一文中研究指出:长期以来关于TLR信号通路中的Toll样受体及其接头分子的起源问题一直都备受关注。七鳃鳗作为现存的最原始的脊椎动物,探究其TLR接头分子的组成与功能特点对探究TLR信号通路的起源进化至关重要。本研究首先基于现有的海七鳃鳗基因组数据库,挖掘鉴定了4个海七鳃鳗TLR接头分子,分别为MyD88、SARM、TICAM-a与TICAM-b。目前日本七鳃鳗MyD88序列已经上传至NCBI,因此本论文以日本七鳃鳗为实验材料,通过分子克隆的实验手段获得了日本七鳃鳗SARM、TICAM-a以及TICAM-b。其全长ORF序列分别为2292bp(764aa)、2373bp(791aa)、1239bp(413aa)。随后对无脊椎动物及脊椎动物各代表性物种TLR接头分子进行了包括系统发育树、结构域组成、序列比对、外显子-内含子基因结构组成分析,以此来探究TLR接头分子的起源与进化。研究发现在漫长的进化过程中,MyD88与SARM序列及结构域组成相当保守。但与MyD88不同的是,SARM的外显子-内含子基因结构在不同的物种中高度变异。我们推测外显子重组(exon shuffling)在SARM多结构域的复杂结构组合和进化中扮演了重要角色。结合系统发育树的分析表明,七鳃鳗TLR接头分子MyD88与SARM都处于无脊椎动物与脊椎动物的交界处,并且其序列结构域及外显子-内含子结构都与脊椎动物更相似,暗示着它们在功能上的保守性,表明二者在进化史上承上启下的重要作用。TICAM分子较为特殊仅存在于脊椎动物中,并且在高等脊椎动物中TICAM分子分化成了TICAM-1以及TICAM-2。本研究发现在海七鳃鳗以及日本七鳃鳗中都含有TICAM-a以及TICAM-b,因此对七鳃鳗TICAM-a、TICAM-b与脊椎动物TICAM-1和TICAM-2的进化关系进行了深度的剖析。首先发现七鳃鳗TICAM-a、TICAM-b在TIR结构域与TICAM-1、TICAM-2十分相似,但无论是TICAM-a还是TICAM-b都不存在TICAM-2中的重要位点。在外显子-内含子基因结构方面,七鳃鳗TICAM-a和TICAM-b都是多外显子,同时TICAM-a的外显子长度总和与TICAM-1的单外显子更为相似,而TICAM-b更像是TICAM-a的复制产物。结合系统发育树的分析,暗示七鳃鳗TICAM-a更有可能为TICAM-1的祖先基因。为了揭示各TLR接头分子的进化历程以及两轮全基因组复制在其进化中的作用,对各接头分子进行了比较基因组学分析。发现在有颌类脊椎动物中MyD88始终与OSR1和SLC22A紧密连接,但七鳃鳗基因组中该区段基因排序高度变异。有颌类脊椎动物基因组中SARM与VTN、SLC46A1紧密连锁,在七鳃鳗基因组中,不存在VTN-SARM-SLC46A1的基因排列,但与斑马鱼SARM所在的基因组区段有其他相同基因,暗示着七鳃鳗与斑马鱼该染色体区段的同源性。由此我们推测,七鳃鳗基因组在第一轮全基因组重复后可能发生了剧烈的染色体重排、同源重组获得丢失等进化事件。最后研究发现七鳃鳗TICAM-a与TICAM-b呈现串联排列,并且其邻近基因并没有与TICAM-1以及TICAM-2相同的FEM1基因。这暗示着TICAM-b是七鳃鳗物种特异性串联重复事件的产物,同时推测TICAM-b在第二轮全基因组重复之前丢失了,而七鳃鳗TICAM-a最终演化为TICAM-1。推测七鳃鳗TICAM-a基因通过逆转录转座作用插入到了另一染色体区段FEM1的附近形成了单外显子结构,并招募了新的5’端侧翼序列最终演变为了TICAM-1。而TICAM-2基因是TICAM-1-FEM1所在的基因组区段经第二轮全基因组重复事件加倍形成的。由此推测第二轮全基因组重复发生在无颌类脊椎动物分化之后及有颌类脊椎动物分化之前。由于脊椎动物中TICAM-1作为TLR3介导的MyD88非依赖途径的重要接头分子,能够响应双链RNA病毒的刺激参与机体的免疫反应。我们通过Q-PCR的检测发现在PolyI:C刺激下Lj-TICAM-a与Lj-TICAM-b能够响应Lj-TLR3在短期4h内的表达上调。但在LPS刺激下则不响应,暗示Lj-TICAM-a、Lj-TICAM-b能够特异性参与双链RNA病毒所诱发的免疫反应。最后本研究通过构建原核表达载体纯化蛋白得到了Lj-TICAM-a和Lj-TICAM-b的多克隆抗体。这些工作都为进一步探究日本七鳃鳗TICAM分子在TLR信号通路中的重要作用奠定了基础。
