作者刘芸芸(2019)在《桦褐孔菌、忧遁草等中药化学成分分离纯化方法研究》一文中研究指出:由于中药所含成分复杂,有效成分含量低,而限制了其利用和发展,因此建立方便有效的分离纯化方法对于促进中药现代化研究及应用尤为重要。本文通过对桦褐孔菌等中草药化学成分分离纯化方法的研究,建立了桦褐孔菌、忧遁草、通关藤、黑老虎及陈皮中化学成分的高效液相色谱(HPLC)分析方法,并采用大孔吸附树脂法(MAR)对其中化学成分的纯化工艺进行了研究,从而建立了方便、高效的纯化方法。1.桦褐孔菌化学成分分离纯化方法研究对桦褐孔菌中的化学成分进行了提取工艺的优化,最佳提取工艺为:用95%乙醇(料液比1:20)回流提取2次,每次0.5 h。采用大孔吸附树脂法对桦褐孔菌三萜类成分的纯化工艺进行研究,以桦褐孔菌中三萜成分的量为指标,比较不同极性大孔树脂对桦褐孔菌三萜的吸附和解吸附性能,然后优化树脂纯化的工艺参数。结果显示,D101型大孔树脂具有较好的吸附和解吸性能,吸附行为更符合Langmuir模型。最佳工艺条件为:5 BV 4 mg·mL-1粗提物溶液上样后,用10 BV 40%乙醇和7 BV 95%乙醇以2 mL·min-1的流速分别进行除杂和纯化,经树脂纯化后的桦褐孔菌三萜含量由25.68%提高至82.00%,确定了本实验所建立的方法能有效纯化桦褐孔菌三萜类物质。2.忧遁草化学成分分离纯化方法研究采用大孔吸附树脂法对忧遁草黄酮类成分的纯化工艺进行研究。以忧遁草黄酮成分的量为指标,比较了不同极性大孔树脂对忧遁草黄酮的吸附和解吸附性能,然后优化树脂的工艺参数。结果显示,HPD-100型大孔树脂具有较好的吸附和解吸性能,吸附行为更符合Langmuir模型。最佳工艺条件为:6 BV 10 mg·mL-1粗提物溶液上样后,用10 BV水和7 BV 40%乙醇以4 mL·min-1的流速分别进行除杂和纯化,经树脂纯化后的忧遁草黄酮含量由61.28%提高至91.74%,确定了本实验所建立的方法能有效纯化忧遁草黄酮类物质。3.通关藤化学成分分离纯化方法研究采用大孔吸附树脂法对通关藤C21甾体皂苷类成分的纯化工艺进行研究。以C21甾体皂苷成分的量为指标,比较了不同极性大孔树脂对通关藤C21甾体皂苷的吸附和解吸附性能,然后优化树脂的工艺参数。结果显示,AB-8型大孔树脂具有较好的吸附和解吸性能,吸附行为更符合Langmuir模型。最佳工艺条件为:8 BV 10 mg·mL-1粗提物溶液上样后,用10 BV 20%乙醇和7 BV 80%乙醇以4 mL·min-1的流速分别进行除杂和纯化,经树脂纯化后的通关藤C21甾体皂苷含量由42.18%提高至88.59%,确定了本实验所建立的方法能有效纯化通关藤C21甾体皂苷类物质。4.黑老虎化学成分分离纯化方法研究采用大孔吸附树脂法对黑老虎化学成分的纯化工艺进行研究,比较了不同极性大孔树脂的吸附和解吸附性能,然后优化树脂的工艺参数。结果显示,HPD-400型大孔树脂具有较好的吸附和解吸性能,吸附行为更符合Langmuir模型。最佳工艺条件为:6 BV 10 mg·mL-1粗提物溶液上样后,用10 BV 20%乙醇、80 BV 40%乙醇和5 BV 95%乙醇以4 mL·min-1的流速分别除去杂质、纯化高极性和低极性成分,经树脂纯化后的黑老虎高极性含量由20.87%提高至88.41%,低极性含量由11.49%提高至81.05%,确定了本实验所建立的方法能有效纯化黑老虎化学成分。5.陈皮化学成分分离纯化方法研究采用大孔吸附树脂法对陈皮黄酮类成分的纯化工艺进行研究。以陈皮黄酮成分的量为指标,比较不同极性的大孔树脂对陈皮黄酮的吸附和解吸附性能,然后优化树脂的工艺参数。结果显示,HPD-400型大孔树脂具有较好的吸附和解吸性能,吸附行为更符合Langmuir模型。最佳工艺条件为:5 BV 80 mg·mL-1粗提物溶液上样后,用10BV水和8 BV 80%乙醇以4 mL·min-1的流速分别进行除杂和纯化,经树脂纯化后的陈皮黄酮含量由42.18%提高至88.59%,确定了本实验所建立的方法能有效纯化陈皮黄酮类物质。
