作者申艳婷(2019)在《水稻病原诱导元件AATCA及其互作转录因子的功能鉴定》一文中研究指出:水稻是世界上重要的粮食作物。由立枯丝核菌(Rhizoctonia solani Kühn)引起的水稻纹枯病是极具破坏性的水稻病害之一。目前水稻病害研究大多集中在白叶枯病和稻瘟病上,很少有针对纹枯病的相关研究。而对水稻纹枯病的抗病机制与调控机理的研究更是少之又少。当前生产上主要是通过培育种植抗病品种来防治作物病害。但由于水稻对纹枯病的抗性是数量性状,所以很难通过常规育种手段来产生高抗纹枯病品种。纹枯病高抗材料的缺乏限制了其抗性育种。因此研究调控抗病相关基因表达的启动子及其互作转录因子进行抗病育种是防治水稻纹枯病的有效策略。前期实验室研究发现,水稻基因Os2H16能够被纹枯病菌所诱导,超量表达Os2H16增强了水稻对纹枯病菌的抗性,抑制表达Os2H16会使水稻对纹枯病菌更加敏感。Os2H16基因启动子是能够被纹枯病菌诱导的病原诱导表达型启动子,并且已经鉴定到了该启动子的主要响应区域为-513 bp至-411 bp以及-411 bp至-309 bp。前期通过烟草瞬时表达系统研究发现一个新的存在于-513 bp至-411 bp区域的能够独立响应纹枯病菌的顺式元件AATCA。本研究通过将AATCA元件转入水稻中进行表达发现其可以响应纹枯病菌的诱导,验证了前期实验室在烟草瞬时表达中的结果。我们通过DNA pull-down技术筛选得到与AATCA元件互作的蛋白。随后运用酵母单杂交技术验证了AATCA元件与OsbHLH057蛋白的互作。通过对OsbHLH057基因进行分析发现其编码一个功能未知的蛋白,具有HLH保守结构域,且定位于细胞核。对OsbHLH057基因的病原诱导表达模式进行分析,发现其可以受纹枯病菌诱导表达,暗示它可能参与水稻对纹枯病菌的响应。然后我们还构建了OsbHLH057基因超量与基因编辑表达载体,获得了水稻超量植株和敲除突变体植株,并且检测了其下游抗病相关基因Os2H16的表达,发现在敲除突变体中该基因表达均有一定程度的下调,表明OsbHLH057基因可能通过影响Os2H16的表达来正调控水稻对纹枯病的抗性。终上所述,本研究对顺式作用元件AATCA及其互作转录因子OsbHLH057进行研究,为利用调控抗病基因的表达,来提高水稻纹枯病抗性的研究提供了新的理论基础,对进一步研究水稻对纹枯病的抗性机制以及培育抗性水稻品种具有重要的意义。
