作者张倩(2019)在《麦长管蚜识别报警信息素EBF的嗅觉蛋白(SaveOBP3/7/9)表达定位》一文中研究指出:麦长管蚜Sitobion avenae Fabricius是小麦植株上重要的刺吸式害虫。嗅觉系统在蚜虫对周围环境的感知、寄主植物定位及产卵地选择等过程中发挥重要作用。外界气味分子穿过触角感器孔道进入淋巴液与气味结合蛋白(OBPs)结合,进而开启下游的信号传递。(E)-β-farnesene(EBF)是麦长管蚜等多种蚜虫报警信息素的主要活性成分,EBF的释放会引起周围蚜虫的忌避行为。与EBF强烈结合的麦长管蚜气味结合蛋白主要是Save OBP3/7/9。目前,上述蛋白在蚜虫触角上的表达位置(感器)尚不清楚。本研究利用扫描电镜对麦长管蚜触角感器超显微结构进行了细致观察;利用whole-mount原位杂交技术结合荧光定量技术观察了Save OBP3/7/9在麦长管蚜羽化后不同时间点的表达定位和表达差异;实现了对Save OBP3/7/9在麦长管蚜成蚜触角上亚细胞水平的表达定位。取得的主要结果如下:1.麦长管蚜成蚜触角感器发育过程的超显微结构观察和Save OBP3/7/9在羽化不同时间节点的表达差异。结果如下:1)成蚜触角感器包括原生感觉圈、次生感觉圈、Ⅰ型和Ⅱ型毛形感器、钟形感器和腔锥状感器等;2)成蚜触角感器的类型及分布相对一致,均分布于触角第3节靠近基部的2/3范围内。数量上,有翅成蚜次生感觉圈的数量(7-13个)则明显多于无翅成蚜(1-5个);3)初羽成蚜触角第3节的次生感觉圈和梗节的腔锥状感器均尚未发育完全,其他感器则形态完整。羽化2h后,麦长管蚜触角与EBF识别相关的次生感觉圈发育完全;4)Save OBP3的表达量在羽化后2h开始上升,这与次生感觉圈数量发育完全是同步的;Save OBP9在麦长管蚜羽化后保持在稳定的较高水平表达;羽化6h后,主要在有翅次生感觉圈表达的Save OBP7表达量上升,该基因表达量动态与次生感觉圈的发育具对应关系,推测其不但参与识别EBF且功能相对专一。2.Save OBP3/7/9基因克隆。根据本实验室获得的Save OBP3/7/9基因全长序列,以Langfang-1为实验种群进行基因克隆。与NCBI数据库中Save OBP3/7/9基因序列比对结果显示,Save OBP7/9序列比对结果完全一致,而Save OBP3可能存在两个拷贝,上述结果为设计并合成Save OBP3/7/9原位杂交探针提供了精准模板。3.SaveOBP3/7/9的触角表达定位。whole-mount原位杂交SaveOBP3/7/9在麦长管蚜有翅和无翅成蚜触角感器上的表达定位结果显示:1)有翅成蚜靶标基因表达部位及感器类型包括触角第6鞭节的Ⅱ型毛形感器、原生感觉圈上的大、小板形感器、第5鞭节原生感觉圈的板形感器、第3鞭节次生感觉圈的板形感器以及触角第3-5鞭节的Ⅰ型毛形感器;2)无翅成蚜靶标基因表达部位主要在触角鞭节。其中,OBP3和OBP9均在触角第6节的Ⅱ型毛形感器、原生感觉圈中的大、小板形感器、第5节原生感觉圈的板形感器、第3节次生感觉圈的板形感器以及触角第4节的Ⅰ型毛形感器上表达;OBP7在触角第6节的Ⅱ型毛形感器、原生感觉圈中的大、小板形感器、第5节的原生感觉圈的板形感器以及触角第4节的Ⅰ型毛形感器上表达。
