作者李继成(2019)在《外侧缰核在调节小鼠厌恶和奖赏行为中的作用及机制》一文中研究指出:奖赏和厌恶系统是调节动物机体“趋利避害”这一动机行为的重要保障,是生物生存、种族延续和生物进化的重要影响因素。奖赏刺激可产生愉悦、兴奋和欢快的感觉,进而导致趋向行为,并最终引导行为加强,诱发生物体有目的、有动机的行为反应,形成一种正性强化作用;相反地,厌恶刺激会导致生物产生恐惧、厌恶和反感等消极情绪,诱发回避行为,表现为一种负性强化效应。外侧缰核(LHb)作为脑内十分重要的负性行为调控中枢,也被称为“反奖赏中枢”,其与脑内重要的奖赏中枢——腹侧被盖区(VTA)之间不仅有着密切的纤维联系,而且二者均可对外界的各种奖赏或厌恶刺激产生反应,参与调节脑内奖赏和厌恶系统。但二者是如何汇聚奖赏和厌恶这两种相反的刺激信息,尤其是其中的单个神经元是否可汇聚并处理厌恶和奖赏这两种不同的传入信息,以及LHb中相关种类神经元对奖赏和厌恶行为的调节作用尚不清楚。在本研究中,结合多通道整体电生理和光遗传技术,我们记录了LHb和VTA中单个神经元分别对奖赏和厌恶刺激这两种不同的刺激信息的反应,分析了奖赏和厌恶信息在单个神经元中的汇聚情况,并进一步对LHb中对奖赏和厌恶刺激均有反应的神经元进行了鉴别,以明确其中谷氨酸神经元所占的比例。之后,结合光遗传技术和行为学检测,分别兴奋或抑制LHb中的谷氨酸神经元,或是VTA中来自LHb的谷氨酸神经纤维末梢,记录观察小鼠奖赏和厌恶相关行为的变化。结果显示,LHb神经元主要对奖赏刺激产生抑制反应,对厌恶刺激产生兴奋反应;而VTA神经元相反,其主要对奖赏刺激产生兴奋反应,对厌恶刺激产生抑制反应。在LHb和VTA中分别有75%和55%的单个神经元可汇聚奖赏和厌恶这两种信息的传入(汇聚神经元),但二者的反应趋势相反:LHb中77%的汇聚神经元为厌恶兴奋-奖赏抑制神经元,且主要是谷氨酸能神经元(88%);而VTA中51%的汇聚神经元是奖赏兴奋-厌恶抑制神经元。更为重要的是,兴奋LHb中的谷氨酸能神经元可明显抑制VTA中的这类汇聚神经元。行为学结果显示,不论是激活小鼠LHb谷氨酸神经元,抑或是激活LHb至VTA的谷氨酸能神经纤维投射均可引发厌恶行为;抑制LHb至VTA的谷氨酸神经纤维投射可产生奖赏行为,然而,抑制LHb谷氨酸神经元却未能诱发奖赏行为。我们的研究表明,厌恶中枢LHb中汇聚奖赏和厌恶这两种不同信息的单个谷氨酸神经元和奖赏中枢VTA中汇聚厌恶和奖赏信息的单个神经元间可能存在相互作用,并在机体奖赏和厌恶行为调节的过程中发挥重要作用。
Abstract
