作者孟畅(2019)在《D~2型细胞质雄性不育小麦的鉴选及育性恢复的评价》一文中研究指出:植物雄性不育是高等植物一种非常普遍的生物学现象,在杂种优势的利用研究中具有重要意义。目前,细胞质雄性不育已成为杂种优势利用的重要手段。为了筛选理想的细胞质雄性不育小麦类型,获得恢复力较强的恢复系和强恢复组合,本研究以本课题组育成的D2型细胞质雄性不育系(Va706A、Ju706A、C6706A)为材料,通过对其进行花药碘化钾染色以及花药、小孢子的扫描电镜观察,明确不育系的败育特征及类型。以K型细胞质雄性不育系(K706A)为对照,将这4种小麦细胞质雄性不育系与本课题组筛选的128个恢复系进行杂交,通过对其F1代进行结实率和农艺性状的测定,比较D2型小麦雄性不育系与K型细胞质雄性不育系在育性恢复方面以及细胞质效应方面的异同点。获得以下试验结果:1.同核异质D2细胞质雄性不育小麦的生物学特征D2型、K型细胞质雄性不育系的花药与恢复系的花药在外形以及内部表型来看,都有明显的区别。恢复系LK783的花药外形挺直,基部开叉,外壁排布比较清晰有规律,小孢子的形状呈圆形,饱满,乌氏体排列紧密。花粉粒染色为均匀的黑色,形状规则、外形饱满,呈圆形。不育系的花药在外形方面形状较短、弯曲,花药的顶端是闭合的不散花粉,花药的外壁排布无序、杂乱无章褶皱,无条理,小孢子的形态不规则,不饱满,呈干瘪状,乌氏体排列稀疏。花粉粒染色不均匀不充分,C6706A属于典败,花粉粒形状不规则,干瘪,呈三角形。K706A、Ju706A、Va706A为染败或者典败。而恢复系LK783的花粉粒染色为均匀的黑色,形状规则、外形饱满,呈圆形。2.D2型细胞质雄性不育系的易恢性的测定筛选K型雄性不育系的F1平均结实率高于D2型细胞质雄性不育系。其中D2型细胞质雄性不育系:Ju706A的F1平均结实率最高,与父本223H2杂交后的结实率高达93.67%,其次是Va706A,其与R354-1杂交后的结实率为91.75%。平均结实率最低的是C6706A的F1,其与陕农33-1杂交结实率最高,为76.97%。从不育系恢复方面的稳定性来看,恢复能力最稳定的为C6706A,Ju706A的稳定性最差,居于中间位置是Va706A。由此可得出结论,相对于K型不育系而言,D2型细胞质雄性不育系其恢复稳定性均低于K型细胞质雄性不育系,易恢性表现为Ju706A>Va706A>C6706A,恢复稳定性表现为C6706A>Va706A>Ju706A。3.恢复系的筛选及评价通过恢复性异地鉴定,恢复系79107对四种不育系的平均结实率于三原、杨凌都是最高的,分别为75.14%、65.02%,该恢复系恢复能力比较强,也比较稳定,能较好的恢复不育系的育性。恢复系K460、A731/L783恢复能力也比较强,对于四个不育系的平均结实率分别为72.52%、75.51%。根据农艺性状(株高)与恢复能力综合筛选,两个恢复系79107、A731/L783可以恢复所有的不育系,有20个恢复系可以单独恢复K706A,有6个恢复系可以单独恢复Va706A,单独恢复Ju706A和C6706A的分别有4个。
