作者崔娜(2019)在《微藻降解废水中间甲酚的研究及应用》一文中研究指出:间甲酚作为苯酚的甲基化衍生物,是酚类化合物中的代表性物质,因具有较高毒性和腐蚀性被列为我国优先控制污染物之一。间甲酚比苯酚具有更高的毒性,在较低浓度时便可对人类和环境造成危害。利用微藻去除废水中的酚类化合物具有快速、稳定、效率高、能耗低等优点,它可以通过生物富集、生物降解等方式将废水中的酚类化合物或其他难降解有机物转化为无毒物质,从而达到化工工业废水的排放要求。本课题以人工配制的间甲酚模拟废水为研究对象,研究了间甲酚生物降解的优势藻种及初始藻密度、初始培养基pH值和初始间甲酚浓度对微藻生物降解间甲酚的影响;比较了蛋白核小球藻驯化前后对间甲酚耐受性和去除效率的变化;探究了蛋白核小球藻对间甲酚的代谢途径;并设计、搭建了藻-膜反应器,考察了间甲酚进水浓度和水力停留时间对反应器运行的影响。得出结论如下:(1)选取对酚类物质有较好降解能力的四种微藻:铜绿微囊藻、蛋白核小球藻、斜生栅藻和鱼腥藻,分别对间甲酚进行生物降解,观察四种微藻对间甲酚的耐受性及降解能力,发现蛋白核小球藻对间甲酚的降解效率最佳;探讨了不同初始藻密度(OD600=0.2、0.4、0.6、0.8、1.0)、不同初始pH值(pH=5.0、6.0、7.0、8.0、9.0)及不同初始间甲酚浓度(100 mg/L、200 mg/L、300 mg/L和400 mg/L)下蛋白核小球藻的生长状况及其对间甲酚的去除效率,发现当蛋白核小球藻的初始藻密度OD600=0.8时,间甲酚的存在对蛋白核小球藻的生长具有明显的促进作用,且4 d内便可将100 mg/L的间甲酚完全降解;培养基初始pH=7.0时,第6 d藻密度为初始藻密度的1.79倍,对间甲酚的去除率达到100%,而酸性条件下的间甲酚去除率仅为28.08%;间甲酚初始浓度对蛋白核小球藻的生长有很大的影响,间甲酚初始浓度越高,对蛋白核小球藻的毒性作用越强,蛋白核小球藻生长及对间甲酚降解的适应期越长。通过一系列的实验,最终确定蛋白核小球藻降解间甲酚的最适初始藻密度为OD600=0.8,最适培养基初始pH=7.0,最适间甲酚初始浓度为100 mg/L。(2)研究了藻种驯化对蛋白核小球藻的生长及对间甲酚降解能力的影响。结果表明,驯化后蛋白核小球藻的适应期缩短,藻密度和叶绿素a含量明显提高,300 mg/L和400 mg/L间甲酚浓度胁迫下的藻体叶绿素a含量分别是未驯化藻体的1.98倍和3.98倍;驯化后藻体对间甲酚的耐受性增加,间甲酚的最大致死浓度由500 mg/L上升到了600mg/L;驯化后蛋白核小球藻在35天内可完全降解400 mg/L的间甲酚,表明藻种驯化可以有效提高微藻的降解能力。(3)探讨了蛋白核小球藻对间甲酚的代谢途径,利用FT-IR和GC-MS的定性分析功能,检测到间甲酚代谢过程中有3-羟基苯甲酸和龙胆酸等中间产物的生成,推测间甲酚可能是通过龙胆酸途径进行降解;间甲酚首先在苯酚羟化酶的作用下代谢形成3-羟基苯甲酸,然后进入龙胆酸途径进一步裂解,最终形成丙酮酸或延胡索酸进入糖酵解(Glycolysis)和三羧酸循环(TCA)。(4)设计、搭建了藻-膜反应器并对间甲酚进水浓度及水力停留时间对藻-膜反应器处理效果的影响进行了研究。结果表明,间甲酚及COD的去除效率随间甲酚进水浓度的升高而降低,随水力停留时间(HRT)的延长而增加。间甲酚进水浓度低于200 mg/L或水力停留时间大于等于3 d时,间甲酚及COD的去除率均可达到90%以上。说明所设计、搭建的藻-膜反应器对间甲酚废水具有较高的去除效率。对反应过后膜丝的污染状况进行了表征,发现反应器内废水及微藻颗粒仅对膜丝表面有轻微污染,对膜孔内部几乎没有影响。
