论文摘要
本研究本实验将丙交酯(LA)、乙交酯(GA)、聚乙二醇(PEG)在0.05%辛酸亚锡催化下共同反应合成PLGA-PEG。通过核磁共振波谱仪证明合成成功。应用XSS-300型熔体挤出机,将PLGA-PEG加工成管状支架,环氧乙烷熏蒸消毒。将血管平滑肌细胞培养在PLGA,PLGA-PEG模片上,应用MTT法及流式细胞仪通过观察平滑肌细胞形态学改变、细胞生长过程的变化,验证材料的细胞相容性。应用酶谱法及Western Blot方法研究PLGA-PEG支架置入对基质金属蛋白酶及C反应蛋白的影响。结果显示:合成的PLGA -PEG细胞相容性好,但以其为材质制备的支架在置入后可触发炎性反应。
