作者聂晨曦(2019)在《青稞β-葡聚糖理化性质及其对肠道菌群的影响》一文中研究指出:β-葡聚糖是一种不能被人胃肠酶消化的可溶性膳食纤维,具有重要的生理功能,尤其是可通过调节肠道菌群结构和功能对宿主健康产生重要影响。藏青25是高β-葡聚糖的西藏青稞品种,其含量可达8.62%。本试验以藏青25为原料,采用碱提醇沉法制备青稞β-葡聚糖,经过DEAE-Sepharose Fast Flow和Sepharose 6B纯化后,分析其纯度、分子量、溶解度、黏度等理化性质。并进一步分析单糖组成、核磁共振、傅里叶红外光谱、原子力显微镜、场发射扫描电镜研究青稞β-葡聚糖基本化学结构。再此基础上,采用体外粪便发酵法、16S rDNA高通量测序技术以及高效气相色谱法研究青稞β-葡聚糖肠道菌群结构和功能的影响。研究结果如下:(1)经碱提醇沉方法制备的青稞β-葡聚糖粗品纯度可达68.35%,再经过DEAE-Sepharose Fast Flow和Sepharose 6B纯化后多糖纯度可达90.61%,可以满足后续试验的纯度要求,同时分子量测定结果为单峰且峰形对称说明其分子量分布均一,分子量为34.456 KDa。溶解度为41.26%,黏度为1.01 mm2/s。(2)青稞β-葡聚糖主要由葡萄糖组成,且葡萄糖通过β-1,3-Glup和β-1,4-Glup链接方式构成糖链。青稞β-葡聚糖固体微粒表面光滑,呈片状、长柱状、椭圆形球状,在溶液中存在一定交联。(3)16S rDNA测序结构发现青稞β-葡聚糖体外发酵液中与产丁酸、丙酸有关的拟杆菌门和厚壁菌门所占比例升高。在科水平上普雷沃氏菌科、双歧杆菌科所占比例上升,肠杆菌科氨基酸球菌科所占比例下降。在属水平普氏菌属9、拟杆菌属、巨球型菌属、巨单胞菌属、双歧杆菌属占比上升,大肠杆菌-志贺氏菌、粪杆菌属、肠杆菌属和Lachnoclostridium占比下降。经KEGG代谢途径的差异分析表明体外发酵空白组与青稞β-葡聚糖组涉及的代谢通路共有24个,其中心血管疾病和病毒性传染病的代谢通路降低。体外发酵试验结果表明,青稞β-葡聚糖具有良好的发酵性,与空白组相比,青稞β-葡聚糖组体外发酵24 h后发酵液中SCFAs含量由高到低依次为:丁酸>丙酸>乙酸,分别是空白组的2.53倍、1.53倍和1.21倍。发酵24 h后空白组pH从7.04下降到6.48,青稞β-葡聚糖组pH下降到4.51。
