目的:通过制作肝缺血再灌注损伤的模型,探讨舒血宁注射液是否对肝缺血再灌注损伤具有保护作用,为临床治疗提供实验依据。方法:取体重为290g-320g的雄性Wistar大鼠50只,随机等分为5组:分别为假手术组,缺血再灌注组,舒血宁注射液小剂量组(30mg/kg),舒血宁注射液中剂量组(60mg/kg),舒血宁注射液大剂量组(120mg/kg)。除假手术组外所有大鼠开腹后游离门静脉和肝固有动脉,用无损伤动脉夹给予钳夹,30min后再灌注120min,形成全肝缺血再灌注模型,假手术组开腹后只行游离肝十二指肠韧带,不行夹闭处理。采集标本后分别进行以下检测:1.血清中的门冬氨酸氨基转移酶(asparta te aminotransferase, AST)和丙氨酸氨基转移酶(alanine aminotransferase,ALT)的水平,超氧化物歧化酶(superoxide dismutase, SOD)的活性以及丙二醛(maleic dialdehyde, MDA)的含量;2.肝组织中的超氧化物歧化酶(superox ide dismutase, SOD)的活性,丙二醛(maleic dialdehyde, MDA)和谷胱甘肽(glutathione GSH)的含量。3.肝组织中的B细胞淋巴瘤/白血病-2(B-cell lymphoma/leukemia-2, Bcl-2)、Bcl-2目关X蛋白(Bax)蛋白的表达。结果:舒血宁注射液小,中,大剂量组较缺血再灌注组:1.均明显降低血清中ALT和AST的水平(P<0.01);2.均明显提高血清中SOD的活性(P(0.01),降低MDA的含量(P<0.01);3.均明显提高肝组织中SOD的活性(P<0.05,P<0.01),降低MDA的含量(P<0.01);4.均明显提高肝组织中GSH的含量(P<0.01);5.均明显增加肝组织中Bcl-2蛋白的表达(P<0.01),同时减少肝组织中Bax蛋白的表达(P<0.01)。结论:舒血宁注射液可有效减轻肝损伤,增强自由基清除能力,增加Bcl-2蛋白的表达和减少Bax蛋白的表达,对肝缺血再灌注损伤具有明显的保护作用。
