作者周升妹(2019)在《玉米赤霉烯酮对断奶仔猪肠道相关基因表达的影响》一文中研究指出:玉米赤霉烯酮(ZEA)污染广泛,危害严重,如何有效防控、减少损害是饲料工业乃至畜牧业亟待解决的科学难题。肠道是吸收、代谢和降解ZEA最主要的器官。肠道的黏膜比身体其他部位更多地接触毒素,最易受ZEA及其代谢产物的毒性,因此探讨ZEA对肠道的影响,同时验证专用复合酶的应用效果。本试验旨在研究低剂量ZEA对断奶仔猪肠道屏障防御、氧化应激和养分吸收功能影响。选取30头健康三元杂交(杜×长×大)断奶仔猪(平均体重14.13±0.80 kg),随机分为3个处理,各组间初始体重差异不显著(P>0.05)。对照组饲喂基础饲粮,ZEA1.0组的饲粮是在基础饲粮的基础上添加1 mg/kg ZEA,ZEA1.0+M组饲粮则是在试验1组的基础上添加2 000 mg/kg的专用复合酶。预试期10 d,正试期35 d。结果表明:(1)ZEA1.0组和ZEA1.0+M组的ADG显著高于对照组(P<0.05)。(2)ZEA1.0组显著降低十二指肠和回肠绒毛的绒毛高度和隐窝深度(P<0.01),显著增加空肠的绒毛高度和隐窝深度(P<0.01),显著升高空肠和回肠的绒毛高度/隐窝深度(P<0.05)。(3)与对照组相比,ZEA1.0组十二指肠绒毛损伤,上皮脱落,小肠腺稀疏,十二指肠腺组织变得疏松;空肠绒毛顶端偶见损伤,小肠腺稀疏,形状变短;回肠中集合淋巴结着色较浅,数目增多。与ZEA1.0组相比,ZEA1.0+M组十二指肠绒毛上皮完整,损伤得到修复;空肠中小肠腺密集;回肠淋巴结着色较深。(4)与对照组相比,ZEA1.0组claudin1的mRNA表达量和蛋白表达量在十二指肠显著降低(P<0.01),在回肠显著升高(P<0.01);occludin的mRNA表达量在十二指肠、空肠显著降低(P<0.01),在回肠中显著升高(P<0.01);小肠以及肠系膜淋巴结Nrf2的免疫阳性反应增强,mRNA表达量和蛋白表达量显著升高(P<0.01);CAT1的mRNA表达量和蛋白表达量在空肠、回肠显著降低(P<0.01);SGLT1的mRNA表达量和蛋白表达量显著升高(P<0.05)。以上结果表明,(1)饲粮中添加1 mg/kg的ZEA能够提高断奶仔猪的日增重,降低料重比。(2)饲粮中添加1 mg/kg的ZEA能够影响断奶仔猪小肠屏障功能,十二指肠有破坏作用,对回肠有促进作用。(3)饲粮中添加1 mg/kg的ZEA能够刺激小肠黏膜以及肠系膜淋巴结,产生氧化应激反应。(4)饲粮中添加1 mg/kg的ZEA能促进断奶仔猪小肠对糖的转运,抑制空肠和回肠对氨基酸的转运。(5)专用复合酶能有效缓解ZEA的破坏作用。
Abstract
