作者周爽(2019)在《基于萘酰亚胺-吲哚荧光母体活性硫探针的设计合成及其性质研究》一文中研究指出:荧光探针具有高的灵敏度和优良的选择性,被广泛应用于荧光传感和医疗诊断等生命科学研究领域,因此开发出性能优越的新型荧光探针具有深远的意义。由萘酰亚胺和吲哚杂化的荧光染料TPFC4具有较长的发射波长(红色荧光)和较大的Stokes位移(大于100 nm),是构建荧光探针优选的母体结构。本文以染料TPFC4为母体,设计合成了三种活性硫荧光探针,对苯硫酚特异性识别的荧光探针TPFC-DNB,能区分检测巯基氨基酸与非巯基氨基酸的荧光探针TPFC-DNBS,可以区分GSH与Cys/Hcy的荧光探针TPFC-SePh。具体内容如下:(1)以2,4-二硝基苯基(DNB)为苯硫酚的识别基团,与荧光染料TPFC4通过醚键连接构建出探针TPFC-DNB。由于识别基团与荧光母体间的光诱导电子转移(PET)过程,使探针的荧光淬灭。当苯硫酚与探针反应时,发生亲核取代反应,识别基团离去,PET过程受到抑制,荧光得到恢复。探针TPFC-DNB在应用过程中表现出了大Stokes位移(140 nm),红光发射(590 nm),优异的选择性和较低的检出限(36 nM)以及快速的响应,并且成功应用于活细胞中苯硫酚的检测。(2)以2,4-二硝基苯磺酰基(DNBS)为识别基团构建了荧光探针TPFC-DNBS,由于识别基团与染料TPFC4之间的PET过程而使探针无荧光。在与巯基氨基酸(Cys、Hcy和GSH)的作用下,磺酰酯键断裂,释放出染料TPFC4,发射出强烈荧光信号。探针TPFC-DNBS的发射波长在590 nm,Stokes位移为137nm,并且能成功检测活细胞内源性和外源性的巯基氨基酸。(3)以苯硒醚为识别基团,结合香豆素与TPFC4构建了荧光探针TPFC-SePh,该探针能够将GSH与Cys、Hcy及其他氨基酸区分开。GSH与识别基团发生亲核取代反应,苯硒基的部分离去,产生强烈荧光信号;Cys/Hcy与探针反应,识别基团离去后产物继续发生分子内重排反应,致使发射出弱的荧光信号。该探针检测过程中的发射波长为570 nm,Stokes位移为120 nm,表现出高选择性和强抗干扰性。
