端粒酶在年龄相关性白内障晶状体上皮细胞中的表达研究

背景和目的 年龄相关性白内障(age-related cataract,ARC),即老年性白内障(senile cataract),其患病率随年龄的增长而明显增高,是世界范围内重要的致盲眼病。目前年龄相关性白内障的发病机制仍不清楚。 端粒(telomere)是染色体末端的重复核苷酸序列,随着细胞的分裂而逐渐缩短,细胞趋于死亡。端粒酶(telomerase)是一种能催化合成端粒序列的核糖核蛋白酶,使细胞不会因端粒耗尽而出现程序性死亡,被认为与肿瘤的发生、细胞的永生化、增生和衰老密切相关。端粒酶由3个亚单位组成,其中端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)被认为与端粒酶活性水平显著相关。 近年来,Colitz等发现实验动物的晶状体上皮细胞(lens epithelial cells,LEC)中表达端粒酶活性,认为端粒酶激活是对氧化损伤、细胞增生或其它未知因素的反应。但端粒酶在人LEC中的表达情况及其与年龄相关性白内障发病之间的关系,国内外均未见报道。 本实验从蛋白和分子水平系统研究了hTERT在不同类型年龄相关性白内障及正常人LEC中的表达,并选用能良好反映细胞增生状态的指标—增殖细胞核抗原(proliferating cell nuclear antigen,PCNA),探讨hTERT和PCNA在年龄相关性白内障LEC中表达的意义和相互关系;运用透射电镜技术对年龄相关性白
