作者张慧静(2019)在《制干辣椒种质资源鉴定及遗传多样性分析》一文中研究指出:基于形态学和SSR分子标记法研究制干辣椒种质资源的遗传多样性,明确不同种质资源之间的亲缘远近关系,并对制干辣椒种质资源的抗逆性和品质性状进行综合评价,为辣椒新品种选育提供理论基础。以新疆生产上推广应用的22份制干辣椒种质资源为材料,对辣椒30个表型性状的变异性、主成分和多样性进行综合分析;通过形态学法和SSR分子标记法,分别对22份制干辣椒品种进行聚类分析并对两种方法的聚类结果进行Mantel相关分析。分别利用18℃低温胁迫和150 mmol.L-1的Nacl溶液胁迫测定种子的相对发芽势,发芽率,发芽指数和相对隶属函数值,综合评价22份制干辣椒品种在种子萌发过程中的耐冷性和耐盐性。测定制干辣椒的辣椒红素以及辣椒素等品质性状,对其进行综合评价。主要研究结果如下:1.22份制干辣椒种质资源具有丰富的遗传多样性。30个表型性状的平均变异系数为26.44%,其中单果干重,单果鲜重和单果数的变异系数最大。平均多样性信息指数为3.05;干辣椒皮和千粒重的平均信息指数最大,约为46.19%;从22份制干辣椒的主成分分析中提取的前7个主成分共贡献了86.778%,其中第一主成分贡献了21.036%。主要包括单果干重,单果鲜重,果肩形状和枝型。通过SSR分子标记筛选的12对引物共检测到86个等位基因,平均为7.166。其中引物CAMS855检测到的等位位点最多(13个);对22份制干辣椒的聚类分析中,30个表型性状分成了三类,SSR分子标记分成了七类,其中表型性状聚类分析中的第二类种质资源和第三类种质资源分别和SSR分子标记聚类分析中的第六类种质资源和第七类种质资源完全相同。2.通过计算耐低温指标的隶属函数值对不同制干辣椒品种进行综合评价,并通过聚类分析将不同制干辣椒种质资源抗逆性分为3个等级。强耐冷性辣椒品种依次为,陕早红>砻丰源9号>砻丰源红冠;这3份材料为制干辣椒抗逆性育种提供了优异的种质资源。3.通过计算耐盐指标的隶属函数值对不同制干辣椒品种进行综合评价,并通过聚类分析将不同制干辣椒种质资源抗逆性分为3个等级。耐盐性强的辣椒品种依次为,砻丰源9号>陕早红>砻丰源红冠;这3份材料为制干辣椒抗逆性育种提供了优异的种质资源。4.对参试材料的果实发育、外观性状和品质性状进行比较,结果表明:辣椒素总量最高的是陕早红,辣椒红色素含量最高的是红艳13号F1。在果实外观品种方面,航椒4号F1的单果鲜重最高,但其果实失水率也是最高的。新选8819的单株鲜重是最高的,其辣椒素和辣椒总量也较高。以上结果表明,30个表型性状通过聚类分析将22份种质资源分成了三类,SSR分子标记将种质资源分成了七类;抗逆性评价中抗逆性较强的品种为陕早红、砻丰源9号和砻丰源红冠;辣椒素总量最高的是陕早红,辣椒红色素含量最高的是红艳13号F1。
